    Effect of tongfu granules on intestinal microflora barrier function in rats with antibiotic-associated colitis

    Objective To investigate the effects of Tongfu granules on intestinal microflora barrier function in rats with antibiotic-associated colitis. Tongfu granules which is a kind of Chinese traditional medicine. The present study aimed to investigate protecting effect of the medicine on impaired intestinal mucosa, and further study effects of Dahuang and Houpo which are the two main components of Tongfu granules. Methods Eighty male rats were divided into ten groups. After making the models of Lincomycin induced antibiotic-associated colitis, blood samples of rats were taken for measuring endotoxin.The colonic contents of rats were collected to culture four floras: bacillus acidi lactici, bacillus bifidus, enterococci bacillus coli. The pathological change of intestinal mucosa was observed using light microscope. Results The body weight of the model group was significantly decreased, a significant difference was shown between Houpo and Tongfu granule in weight. The endotoxin in rats of treatment group was decreased. But insignificant difference was shown among Dahuang, Houpo and Tongfu granule groups in endotoxin. There was a significant difference (P<0.05) between Tongfu granule and model group after treatment. The number of bacillus acidi lactici, bacillus bifidus, enterococci bacillus coli increased. The scores of the lesions showed a significant difference between Tongfu granule and Dahuang. Conclusion There was injury in intestinal mucosal barrier in antibiotic-associated colitis rats. Chinese Traditional medicine Tongfu granules Dahuang and Houpo had protective effects on the smalI intestinal barrier. They can decrease the endotoxin level of antibiotic-associated colitis rats. They also could recover the intestinal flora. Tongfu granule was better than Dahuang and Houpo.

    Comparison between Different Methods to Extract Mitochondria and Its Effects on Mitochondria Content and Activity

    Objective To compare different methods for extracting mitochondria and its effects on mitochondria content and their activity. Methods Pulilai kit, sucrose gradient centrifugation and Pierce mitochondria extraction kit were used to extract mitochondria and quantify their concentration, activity and contamination. Results Pulilai kit yielded higher concentration of mitochondria proteins than two other methods(P<0.05) when the same amount of cells were used. Western blotting was used to evaluate the amount of cytosolic protein contaminant and mitochondria protein cytochrome c from mitochondria fraction. The result suggests that using Pulilai kit can yield lower concentration cytosolic protein contaminant and higher concentration of cytochrome c from mitochondria fraction than the other two methods. At the same time, ATP content of mitochondria extracted by the method of Pulilai kit was the greatest. Conclusion Pulilai kit is better than sucrose gradient centrifugation and Pierce mitochondria extraction kit for mitochondria extraction.

    Objective The study was designed to investigate the clinical characteristics of aspirin resistance in patients with cardiovascular diseases.Methods 326 patients with stable cardiovascular diseases were evaluated for aspirin resistance with idetection of optical platelet aggregation induced by arachidonic acid(AA) and adenosine diphosphate(ADP). Aspirin resistance was defined as an average aggregation of over 20% with 0.5 mmol/L AAand over 70% with 10 μmol/L ADP. Aspirin semiresistance met one, but not both of the above criteria. Results 4.9% of the patients were aspirin resistant and 27.6% were aspirin-semiresistant. The clinical characteristics of aspirin resistant patients compared with aspirin sensitive patients were: 1) most of the aspirin-resistant patients were senile; 2) female patients were more frequent than male; 3) patients with diabetes were dominant in number; 4) it had nothing to do with hypertension, hyperlipodemia, red blood cell count, hemoglobin and platelet count; 5) it was relatively rare in smoking population. Conclusion The incidence of aspirin resistance is similar to other studies, in addition to aging and female gender, diabetes is also a risk factor in aspirin resistance.

    Lack of the naturally occurred psoriasis animal models once made the researches on human psoriasis phenotype and pathogenesis stagnated in last several years. But with the increasing understanding of psoriasis mechanism, a series of models has been explored at organic, cellular and molecular levels. We evaluated the current psoriasis models and analyze theirs advantages and limitations, to provide reference for psoriasis studying, drug screening and treatment.

    Comparison of Three Imaging Methods in the Early Diagnosis of the Breast Cancer

    Objective To study the value of 3 different imaging methods in the early diagnosis of breast cancer and differential diagnosis of benign versus malignant nature of breast tumors. Methods Three hundred and ten patients with breast disease seen from 2004 to 2008 were randomly selected in this study. Diagnosis of all the patients were confirmed by means of surgery and pathological examinations, and all the patients were examined by high frequency ultrasound and mammography before operation. Using the results of pathologic examination as the gold standard, the authors compared the result of high frequency ultrasound with that of mammography, and compared the result of any one method with the combined results of the other two. The authors also reviewed the results of 20 patients with breast carcinoma in whom magnetic resonance imaging was additionally applied to see its value in the diagnosis on breast carcinoma. Results The rate of consistency of the ultrasound on diagnosing breast carcinoma was 85 percent, while that of breast benign disease was 89.5 percent. The rate of consistency of the mammography on diagnosing breast carcinoma was 86 percent, while that of breast benign disease was 83.8 percent. The ultrasound could identify 85 of the 100 cases with breast carcinoma. While the mammography could make diagnosis for 86 of 100 patients with breast carcinoma. The combination of the both could make diagnosis for 95 of the 100 patients with breast carcinoma. In the 20 patients with breast carcinoma who were examined by magnetic resonance imaging, the accordance rate for diagnosis with the combination of the three was 100 percent. Though no significant difference in the diagnostic accuracy was found between the ultrasound and the mammography, the combination of the both could greatly elevate the diagnosis accuracy. Conclusion 1 The combination of mammography and high frequency ultrasound can be the first choice of methods to diagnose breast carcinoma in the early stage; 2 MRI can be a good supplement to mammography and high frequency ultrasound; 3 Mammography has great importance in diagnosing breast precancerosis-breast atypical hyperplasia, it is a much better method to diagnose breast atypical hyperplasia.

    Correlation between the counting results of Durham pollen sampler and automatically pollen monitor

    Objective To analyze the correlation between the counting results of Durham pollen sampler and automatic pollen monitor, discuss the advantages and defects of two instruments. Methods Durham sampler and KH3000-1 automatic pollen monitor were set up next to each other on the same altitude. Airborne pollen concentrations were measured at the same time. Results Durham sampler has the merit of low cost and can classify the granules that adhere to the sampling surface, but pollen counting using Durham sampler require much time and skilled labor; KH3000-1 automatic pollen monitor count pollen particle using laser light in real time for computer processing, without the counting using a microscope, but it is difficult to distinguish pollen species. Conclusion The onset and peak time of pollen concentration measured by two devices were consistent, the results of automatic pollen monitor affected by pollens particle size and meteorological factors.

    Adoptive transfer of murine bone-marrow derived macrophages tracing in vivo and application in hypertensive cardiac injury

    Objective Two types of mice enhanced green fluorescent protein (EGFP) transgenic mice and CD45 type I (CD45.1) mice) to observe whether the adoptive transfusion of murine bone-marrow derived macrophages(BMM)might affect the hypertensive damage to heart in murine hypertension model. Methods After obtaining bone marrow cells from EGFP transgenic mice and inducing cells with macrophage colony-stimulating factor (M-CSF) to BMM, wild-type mice received injection of 2×106 EGFP+BMMs or phosphate-buffered saline (PBS) through tail vein and observed survival proportion of EGFP+cells in peripheral blood at 1st day and 7th day. Then we collected heart, liver and spleen to determine expression of GFP+ cells. Likewise we used CD45.1 BMM to inject into CD45.2 mice and determine the proportion of CD45.1+ cells in peripheral blood. Then angiotensin Ⅱ (Ang Ⅱ) was infused to the mice and EGFP+BMMs or PBS was infused as previously described. After 7 days infiltration of EGFP+BMM, inflammation and fibrosis in heart were determined.Results The proportion of EGFP+cells in peripheral blood in EGFP+BMMs injection group were 1st day:(4.6±0.5)%; 7th day:(4.4±0.5)% while none in control group. After 7 days GFP+cells expressed in spleen while none in liver, kidney or heart. The proportion of CD45.1+cells in peripheral blood in CD45.1+BMMs injection group was (4.3±0.4)% while none in control group. In contrast to control group, AngⅡ infused group had accelerated increased blood pressure, infiltration of inflammatory cells and collagen deposition. Injection of EGFP+BMMs could infiltrate in heart and accelerate inflammation and fibrosis, but did not affect blood pressure. Conclusion This study proved that adoptive transfuse BMMs could survive in recipient and accelerate cardiac fibrosis. We settled the problem of tracing the adoptive transfused BMM in vivo.

    Evidence-based Medicine Approach and TCM Therapeutic Evaluation

    WHO promotes evidence-based traditional medicine. Methodological issues limit evaluation of therapeutic effect of TCM. Current methods for evaluation do not properly reflect the humanity characteristics and holistic approach and differentiation of syndrome and prescription. TCM therapeutics is a complex intervention, and there is a limitation of methodology by only randomized, double-blind, placebo-controlled trials as it doesn't comply with bio-psycho-social-environmental and spiritual model for modern medicine. To fairly evaluate TCM therapeutic, we need to combine both quantitative and qualitative methods together, so as to create modality and methodology for TCM differentiation of syndrome and treatment.

    Establishment of Animal Model of Human Cervical Cancer in BALB/c Nude Mice by Tumor Tissue Xenograft

    Objective To establish the human cervical cancer animal model through abdominal subcutaneous transplantation in BALB/c nude mice and investigate the rate of tumor-forming and the biological features of the tumor.Methods Human cervical cancer tissue derived from surgical specimens were transplanted into the subcutis of nude mice.The rate of tumor transplantation,generic characteristic,tumor growth of bearing cancer mice were observed every day.On day 85 the tumor-bearing mice were killed and tumor masses were collected for analysis of histological features.Genomic DNAof tumor tissue,peripheral blood,liver,spleen,kidney,omentum,and uterus annex of tumor-bearing BALB/c nude mice were extracted,and then using all organs' genomic DNAas templates,we detected the HPV DNAexpression with polymerase chain reaction(PCR).Results In this experiment,the delitescence period of tumor-forming were 30~35 days.On day 42 the tumor tubercles were visible and grew fast day by day.The average tumor-take rate was 90.5%,and the mean tumor surface areas was(50.0±5.1)mm2 on day 80.The transplanted tumors grew well by local invasion.Metastasis was not found.The tumors were analyzed by HEstaining,and the histological appearance of the tumors before and after transplantation were very similar.The positive rate of HPV DNAin the tumor tissue was 100%.The appearance of HPV DNAbefore and after transplantation was the same.The PCRresults definitely displayed the subtype of high-risk human papillomavirus,which suggested cervical cancer patient's HPV16 infection,not HPV18 infection.But HPV DNAwas not detectable in peripheral blood,the spleen,the liver,the kidney,the omentum,and the uterus annex of tumor-bearing BALB/c nude mice.Conclusion Human cervical cancer model in nude mice has been successfully established which simulates the biological features of human cervical cancer.The animal model of human cervical cancer in BALB/c nude mice exhibit a high rate of tumor transplantation,and which may provide a convenient and ideal tool for the research of the mechanism and therapy of human cervical cancer.

    On the Reliability and the Validity of a Scale

    The object of the reseach was to describe the effect of the negative questions in a scale on the reliability of the scale and the procedure for recoding the negative questions as well as the hypothesis testing method for the questions in a scale, to investigate and to compare the reliability of a scale obtained with different methods, to compute the parallel reliability between two groups of the positive questions in a scale, to analyze the validity of a scale. The method of the parallel reliability, the method of the parallel reliability between two groups of the positive questions, the indpendent t-test method, the canonical correlation, the factor analysis, the methods of computing the Cronbach's coefficient α, the Theta coefficient θ and the Omega coefficient Ω were used. ① Based on the surveillance data of "The Scale of Training the Rural Doctors in Beijing", the comparison of the Cronbach's coefficient α before and after recoding the negative questions was made. ② By using independent t-test to test the null hypothesis of the critical ratio of every question respectively, two questions were sieved and the Cronbach's coefficient rose from 0.892 9 to 0.899 3 after sieving. ③ The simple parallel reliability(r1=0.602 7)was much less trustworthy than the parallel reliability between two groups of the positive questions(rp=0.832). ④ The following indicates the whole scale, α =0.899 3, θ = 0.881 3, Ω =0.956 5 and rp=0.823 0 which showed the scale had excellent reliability. ⑤ For the convenience of discussion, we used one model of the scale named "the reasons of the rural doctors to stay in village or to leave from village" to obtain a distinct statistic structure with the factor analysis method, which showed that the questions in the model could be divided into two groups: one for practical questions, the other rational questions. Endpoints: ① It is important to recode the negative questions in the scale, to compute the reliability and the validity of the scale after sieving the questions without significance. ② It is trustworthy to assess the reliability of the scale with α, θ, Ω and the parallel reliability between two groups of the positive questions except the simple parallel reliability. ③ The statistical structure of the scale obtained by the factor analysis has a very distinct structure, but not unique. The explanation of the statistical structure is based not only upon the statistics but also upon the professional knowledge. And the statistical structure can be a clue to modify the scale.

    Hepatotoxicity Caused by Commonly-used Chinese Medicinal Herbs and Compound Preparation

    The manifestations of hepatotoxicity caused by Chinese medicinal herbs are similar with ordinary liver disease. Many pathological alterations of hepatotoxicity can be seen, such as hepatocellular damage, liver disfunction, increased serum transaminase, hepatomegalia, jaundice, cholestasis, chronic hepatitis complicated with fibrosis, hepatic cirrhosis、fulminating hepatic failure, liver tumor, and etc. In most cases, the liver damage is reversible after discontinuation. The commonly-seen herbs which may cause hepatotoxicity are Huang Yao Zi(Tuber Dioscoreae Bulbiferae)、Cang Er Zi(Fructus Xanthii Sibirici)、Lei Gong Teng(Radix Tripterygum)、Ku Lian Zi(Fructus Meliae Azedarach), etc., and the compound preparations are Zhuang Gu Guan JIe Wan(The Pill for Asthritis), Ke Yin Wan(The Pill for Psoriasis), Xiao Chai Hu Tang (Xiao Bupleuri Decoction), and so on. In addition, the improper combination of Western medication and Chinese medicinal herbs may also bring about hepatotoxicity. Except the toxicity of herbs themselves, the other factors include the dosage, time, the body constitution of patients, sensitivity, seasons, and geographic regions, etc. Therefore, the medicinal herbs and their preparations should be used rationally under the guidance of the theory of traditional Chinese medicine and differentiation of symptoms, and the medical skill should be improved, which are the effective measures for avoiding hepatotoxicity.

    Professor Fa-zi Cong has worked as a clinician of Traditional Chinese Medicine(TCM) for more than fifty years and made great achievement in the treatment of luteal phase defect(LPD)-induced infertilitas feminis.In this manuscript,we collected Professor Cong's idea that the LPD-induced infertilitas feminis could be healed by menstrual cycling therapy according to the TCM differentiation of symptoms and diseases.During regulating menstrual cycle of patient by the TCM differentiation of Chinese herb usage,the following key points should be referred:the critical time of menstrual cycle,the basal body temperature(BBT) curve,and the adjustment on meals.

    Epidemiology of Cardiovascular Disease and Risk Factors in Chinese Population

    Cardiovascular diseases (CVD) have become the leading contributor to death in China for the recent 20 years. Alarge number of epidemiologic studies of CVD were carried out in different regions of China. The study summarized the epidemiologic characteristics of CVD risk factors, distribution and the general tendency. The main results were as follows: Beijing area was different from western countries, the characteristics of CVD in Chinese population showed that both the incidence and mortality rate of coronary heart disease were low, and those of cerebrovascular disease were high. There were significant geographic variations in both CVDincidence and mortality rate, with higher rates in north and lower rates in south. In the past several decades, the increasing trend of CVD incidence was observed in Chinese population, while the CVD related mortality dereaded in most developed countries. Althrough total serum cholesterol (TC) and body-mass index (BMI) of Chinese population were lower than those of western countries, the current trends were increased significantly. Smoking rates in Chinese men ranked No. 1 in the WHO-WONICA study. The CVD epidemic in China was attributable to increased trends of main risk factors of cardiovascular diseases. Comprehensive prevention and intervention for main cardiovascular risk factors could reduce the risk of CVD.

    Application of Double Staining of In Situ Hybridization and Immunohistochemistry in Brain Slices

    Objective To investigate the use of immunohistochemistry(IHC) in combination with in situ hybridization(ISH) to detect a special gene at nucleotide level and protein level at the same time on whole brain slices. Methods After culturing the whole brain slices of rats, HE staining was applied to assess the quality of frozen section of brain slices. Then ISH was used in combination with IHC to detect the expression of a special gene. Results Whole brain slices grew well enough to be used in following research. Compared to either IHC or ISH, combination of both was able to detect the gene at nucleotide level and protein level at the same time. Conclusion Double staining was superior to IHC or ISH. This technique could be used in detecting the nucleotide and protein in brain slices.

    Evaluation of Middle Cerebral Artery Occlusion Induced Focal Cerebral Ischemia Model in Mice

    Objective To study the success rate and reliability of middle cerebral artery occlusion(MCAO) model by objective and reliable neurological score, infarct volume, and edema rate. Methods Cerebral ischemia model of MCAO was established in mouse using the suture method. Infarct volume, edema rate and behavioral performance was evaluated by 2,3,5-triphenyltetrazolium chloride(TTC) staining and neurological score. Results MCAO could induce obvious focal cerebral ischemia by TTC staining and the success rate of this model was about 90.8%. Besides, MCAO could induce three types of cerebral ischemia: cortical, hippocampal and thalamic ischemia. And the predominant type is cortical ischemia. MCAO-induced cerebral ischemia could significantly increase the neurological score compared with the sham group. The procedure for infarct volume evaluation of the MCAO model was accurate and stable; infarct volume was about 35% of entire brain volume. Conclusion Cerebral ischemia model of MCAO was established in mice successfully(90.8%). The evaluation of neurological deficit and infarct volume was technically reliable.
