用鸡胚皮肤作绿脓杆菌粘附的定量研究——Ⅱ 粘附机理的探讨

摘要: 用本室建立的绿脓杆菌(PA)对鸡胚皮肤的体外粘附模型,研究了各种不同的预处理对PA粘附的影响。酸处理皮肤能增强PA的粘附作用,而过碘酸钠处理则能抑制PA的粘附,提示粘附受体中含有糖成份。4种单糖(D-半乳糖,D-甘露糖,L-岩藻糖和N-乙酰葡萄糖胺)处理细菌对PA的粘附作用无明显影响,而唾液酸能显著地抑制PA的粘附,提示唾液酸可能是PA的粘附受体成份。神经氨酸酶和PA-Lectin的处理,对PA粘附无任何作用。从Slime菌株S6中提取的SPS能增强S36和S51菌株对皮肤的粘附,其抗血清则能抑制S36和S51菌株的粘附。透射电镜观察显示S菌株体外糖毡包结构。以上结果表明,SPS是一种重要的粘附物质。

Abstract: Using the chick-skin model,we have investigated the effects of various pretreatments of the skins on adherence.Acid treatment enhanced adherence of the two types of P.aeruginosa.The reduction of adherence after sodium metaperiodate treatment suggests that the adherent receptor contains sugar,but the pretreatment by the four sugars(D-(+)galactose,D-(+)mannose,N-acetylglucosamine and L-fucose)without the significant inhibition of P.aeruginosa adherence.Only sialie acid inhibited the adherence of these strains. Galactose-specific P.aeruginosa lectin and CL.perfringens neuraminidase had no inhibitory effect on the adherence of the organisms.SPSpurified from slime strain S36 at concentrations as low as 50μg/ml enhanced adherence of the strain S51 and strain S36 The antisera to SPS inhibited the adherence of strain S36 and S51 but not the adherence of nonslime strain NS 8. Direct electron microscopic examination of slime strain S36 have shown that thestengancymof slime strain of P.aeruginosa to produce a thick, flexible,exopolysaccharide-enclosed glycocalyx.It is evident that SPS is an important material in adherence of P.aeruginosa stralns to the skins.
