Sertoli细胞对体外培养的大脑皮质神经元的营养作用

Trophic Effect of Sertoli Cells on Cerebral Cortex Neurons in Vitro

摘要: 为研究睾丸支持细胞(Sertoli cells,SCs)对体外培养的大脑皮质神经元的营养作用,采用2种实验方法:1)将Sertoli细胞与大脑皮质神经元共培养;2)在神经元培养基内添加Sertoli细胞条件培养基(Sertoli preconditioned medium,SCM)。结果:在培养早期SCs能显著促进大脑皮质神经元的贴壁,对神经元胞体及突起的生长也有显著的效果,培养晚期SCs组的神经元生长状况也显著优于DMEM培养组和SCM组,说明SCs对神经元有显著的促生长作用;同时实验还证实SCM对神经元在体外的生长也具有一定的促进作用。

Abstract: The aim of the present study was to find out whether Sertoli cells(SCs) can provide trophic factors for cerebral cortex neurons in vitro or not. Fetal mice cerebral cortex neurons were cultured alone,cultrured with Sertoli preconditioned medium(SCM) or cocultured with mice Sertoli cells. Results showed that SCs significantly enhanced the viability and growth of cerebral cortex neurons during early days in vitro. During later days, neurons cocultured with Sertoli cells were still better than the other two groups. So it is testified that Sertoli cells can improve the growth of neurons in vitro. SCM in this research also showed undoubted trophic effect on cerebral cortex neurons in vitro.
