乳腺癌中β-catenincyclinD1c-myc表达及意义

Expression and significance of β-catenin, cyclinD1, c-myc in breast carcinoma

摘要: 目的 探讨β-catenincyclinD1c-myc在乳腺癌中的表达及意义。方法 应用免疫组化法检测119例乳腺癌,72例乳腺增生症和40例正常乳腺组织中β-catenincyclinD1c-myc的表达,并分析其与乳腺癌临床病理特征的关系。结果 40例正常乳腺组织中β-catenin在细胞膜中强表达,cyclinD1c-myc阴性表达。乳腺癌β-catenin异常表达率78.15%(93/119)明显高于乳腺增生症44.44%(32/72),组间比较差异有统计学意义(P<0.01)。乳腺癌cyclinD1阳性表达率57.98%(69/119)高于乳腺增生症33.33%(24/72),组间比较差异有统计学意义(P=0.001<0.01)。乳腺癌c-myc阳性表达率56.30%(67/119)高于乳腺增生症27.78%(20/72),组间比较差异有统计学意义(P<0.01)。β-catenin异常表达、cyclinD1的过表达与淋巴结转移相关,c-myc的过表达与肿瘤大小和淋巴结转移相关。β-catenin的异常表达与cyclinD1的表达相关,而与c-myc的表达无关。结论 β-catenin异常表达是乳腺癌发生发展过程中的重要因子,这个过程可能是通过激活cyclinD1实现的。β-catenin异常表达和cyclinD1c-myc过表达可作为评估乳腺癌转移预后的指标。

Abstract: Objective To study the expression and significance of β-catenin, cyclinD1, c-myc in breast carcinoma. Methods Breast tissue specimens from 119 patients with breast carcinomas, 72 patients with breast cystic hyperplasia and 40 normal individuals were studied. Immunohistochemistry was used to detect the expressions of β-catenin, cyclinD1, c-myc, then their relationship with the clinicopathological features of patients with breast carcinoma was analyzed. Results In normal breast tissues, from the 40 control epithelial cells showed equally strong membranous expression of β-catenin protein at the cell-cell boundaries, but the expression of cyclinD1, c-myc was negative. The abnormal rate of β-catenin in breast carcinoma(78.15%) was much higher than that in patients with breast cystic hyperplasia tissues(44.44%), significant correlation was found(P<0.01). The overexpression of cyclinD1, c-myc in breast carcinomas(57.98%, 56.30%) was much higher than that of breast cystic hyperplasia tissues(33.33%, 27.78%), significant correlation was found(P<0.01). The abnormal expression of β-catenin and the overexpression of cyclinD1 was associated with lymph node metastasis(P=0.031, P=0.003). The overexpression of c-myc was associated with size and lymph node metastasis(P=0.041). There was a highly significant association between abnormal expression of β-catenin and overexpression of cyclinD1(P=0.006), but not between abnormal expression of β-catenin and overexpression of c-myc(P=0.775). Conclusion The abnormal expression of β-catenin may play an important role in the carcinogenesis and progression of human breast carcinoma by up-regulating the expression of cyclinD1. β-catenin abnormal expression and the cyclinD1, c-myc overexpression could be considered as useful makers for determining metastasis and prognosis of human breast carcinoma
