用猪血清Clq提取物检测循环性免疫复合物

摘要:

我们采用Yonemasu优球沉淀及Bing亲和层析法,并附加SPA菌体吸收IgG的步骤,从猪血清分离Clq。经热灭活,化学灭活,4%SDS-PAGE分析证明不仅在Clq提取物中而且在高度提纯的猪IgM中均含有IgM-RF。用猪Clq作固相ELIZA,试验的敏感范围是10-1000ngHAG/mL。38例正常人血清的测定值是2.06±1.52μgHAG/Ml(m±SD)。测13例SLE及16例RA病人血清,其值分别为31%及62.5%,超过m+2SD。用猪Clq测CIG可取得满意结果。

Abstract:

Porcine Clq was purified by the euglobulin precipitation method of yonemasu and the affinity chromatography method of Bing. Contaminating IgG was absorbed away with lyophilyzed staph,aureus Cowan I type. Such Clq preparations were shown to contain IgM-RF by heat-inactivation studies and SDS-PAGE analysis. Highly purified porcine IgM had RF actively as well. When it was used in the solid phase Eliza test to detect CIC, the sensitivity range was 10-1000ng HAG/ml. 38 normal sera had values of 2.06±1.52 ng HAG/ml(m±SD). Sera from 13 cases of SLE and 16 cases of RA had positive rates of 31% and 62.5% respectively(2 SD). The satisfactory results performance and cheapness of the pig blood are in favour of the use of porcine Clq in CIC assays.
