摘要: 本文介绍用鸡胚皮肤作绿脓杆菌(PA)粘附的体外模型的方法。无菌操作取9~10日龄鸡胚背部皮肤3块(每块约20mm2)与细菌孵育后用扫描电子显微镜对固定面积的细菌进行直接计数。粘附最适条件:接种菌量为107~108CFU/mL,孵育时间为1小时。结果表明,PA与皮肤能发生明显粘附,有些slime菌株比nonslime菌株的粘附作用高10倍以上。加热或甲醛杀死的细菌不粘附皮肤。PA抗血清也能抑制细菌粘附。其他革兰氏阴性杆菌(大肠杆菌,肺炎杆菌和变形杆菌)对皮肤的粘附作用较弱。这一模型对于研究PA与损伤皮肤所发生的早期感染过程将是十分有用的。

Abstract: A model of the adherence of Pseudomonas aeruginosa to chick skins is described.The skins which were separated from the dorsal part of the embryo were cut up into pieces about 20mm2 and placed in petri-dishes for adherence studies.Adherence was quantification by direct count of the number of bacteria attached to a fixed surface area as viewed by scanning electron microscopy.Optimal condition for study were 1 h of incubation with an inoculum of 107-108CFU/ml.Under this condition,the slime strains adhered to the skins 10 fold better than the nonslime strains.Heat and formalin treated P.aeruginosa strains did not adhere to the skins,and antiserum against P.aerugiosa can inhibit adherence.Bacteria species such as E.coli,Klebsiella pneu-moniae,and Proteus mirabilis adhered poorly to the skins in this system. This model should be useful in studying the reaction of bacteria with the skins of burn patients.
