黄芪对体外人胚肺二倍体成纤维细胞抗衰老作用的研究Ⅰ.细胞寿命、增殖能力和超氧化物歧化酶含量的研究

An Investigation of Anti-aging Effects of Astragalus Membranaceus Bunge on Human Embryonic Lung Diploid Fibroblasts in Vitro ——Ⅰ.A Study of Life-span,Reproduce and SOD Content of Cells

摘要: 以体外培养的人胚肺二倍体成纤维细胞为材料,当细胞培养至27代时,将细胞分为加黄芪提取液的给药组和不加黄芪的对照组,相同条件下传代培养,直至细胞停止增殖。同时,每隔一定代龄用流式细胞计测定两组细胞的增殖能力,以化学发光法测定其 SOD 含量,并进行常规染色体检查和细胞一代存活时间的实验。结果表明:给药组生长77代,而对照组细胞仅存活至52代。至传代终止两组细胞都始终保持其二倍体性质,随着代龄的增加,两组细胞的增殖能力、SOD 含量和一代存活时间均明显下降。同一代龄,给药组细胞的增殖能力、SOD 含量和一代存活时间高于对照组,随代龄的增加,这种差距更加明显。

Abstract: Human embryonic lung diploid fibroblasts(HELDF)in vitro were used as experiment materials.Whe the cells were subcultured for 27 generations,they were divided into two groups,medicine-treated group(B)which was supplemented with Astragalus membranaceus Bunge(AMB),and control group(C).The two groups were subcultured at same condition until the cells ceased proliferating.In the meantime,at regular intervals,the proliferation a-bility and SOD contents of two groups were determined with the flow cytometry and the chemiluminescence method respectively.The routine chromosome test and the measurement of the survival time in one generation were also taken at same time.Our results showed that the cells of group B were subcultured for 77 generations while the cells of group C only for 52 generations.The HELDF of two groups maintained their diploidy from the beginning of subculture to the end.The proliferation ability,SOD contents and the survival time of two groups decrease gradually with the increase of generations.But at the same generation the proliferation ability,SOD contents and the survival time of group B was higher than those of group C and this difference became more obvious with the increase of generations.
