Effects of iNOSmRNA on Pancreatic Endocrine Function of Experimental Severe Acute Pancreatitis in Rats

摘要:

目的 研究重症急性胰腺炎大鼠胰腺内分泌功能的变化,探讨iNOSmRNA在胰腺内分泌功能损伤中的作用。方法 制作重症急性胰腺炎大鼠动物模型,对胰腺病理损伤评分;检测脂肪酶、血糖浓度;分离胰岛,进行葡萄糖刺激胰岛素分泌试验;应用逆转录聚合酶链反应技术,检测胰岛iNOSmRNA的表达情况。结果 重症急性胰腺炎大鼠病理学评分、血糖、血清脂肪酶升高,葡萄糖刺激胰岛素分泌量较对照组明显减少,差异有统计学意义(P<0.05);逆转录聚合酶链反应提示重症急性胰腺炎组胰岛的iNOSmRNA表达增强,差异有统计学意义(P<0.05)。结论 重症急性胰腺炎对胰腺内分泌功能有较大影响,iNOSmRNA在其中发挥作用.

Abstract:

Objective To investigate the endocrine function changes in rats with severe acute pancreatitis, and explore the effects of iNOS on pancreatic endocrine function of experimental severe acute pancreatitis in rats.Methods SAP model was produced and the pathological score and the level of serum glucose and lipases were determined. Islets of rats with SAP were isolated and the amount of insulin secretion stimulated by glucose was determined by radioimmunoassay (RIA). The iNOSmRNA expression in isolated islet cells were assayed by reverse transcription PCR.Results In SAP models, pancreas showed pathological injury after induction of pancreatitis, accompanied with the elevated levels of serum glucose and lipases. The amount of insulin secretion induced by glucose from rats' islets with SAP were less than those from normal rats. In SAP rats, the iNOSmRNA expression of islet increased significantly.Conclusion SAP could impair the pancreatic endocrine function, and iNOSmRNA has been involved in the islet functional injure in SAP.
