野生型p53,GM-CSF和B7-1基因转移对卵巢癌细胞系SKOV-3增生的影响

Effects of Wild-type p53,GM-CSF and B7-1 Genes Transfer on Proliferation of Ovarian Cancer Cell Line SKOV-3

摘要:

目的 研究人野生型p53、GM-CSF和协同刺激因子B7-1基因共转染对卵巢癌细胞增生的影响。方法 以携带绿色荧光蛋白基因的重组腺病毒载体转染卵巢癌细胞系SKOV-3,荧光显微镜观察,流式细胞计数计算转染效率;以携带野生型p53、GM-CSF和B7-1基因的腺病毒转染SKOV-3细胞,RT-PCR检测目的基因表达,观察转染前后细胞形态的变化,绘制细胞生长曲线。结果 当腺病毒的感染强度达到400 pfu/细胞时,转染率可达到81%;以携带目的基因的腺病毒转染SKOV-3细胞,能检测到相应基因表达;转染后SKOV-3细胞形态发生改变,增生减缓。结论 腺病毒载体能介导目的基因转入卵巢癌细胞,并有效表达;人野生型p53、GM-CSF和B7-1基因共转染能减缓卵巢癌细胞的增生。

Abstract:

Objective To investigate the effects of wild-type p53,GM-CSF and B7-1 genes co-transfer on proliferation ability of ovarian cancer cell line SKOV-3.Methods Transfection SKOV-3 cell with recombinant adenovirus carrying green fluorescence protein gene was used to test the transfection efficiency of adenovirus vector.Transfect SKOV-3 cell with recombinant adenovirus carrying human wild-type p53,GM-CSF and B7-1 genes,then RT-PCR was used to test the gene expression.The appearances of the cells after transfection were compared with the normal cells.Proliferation curve was performed.Results The transfection efficiency can reach 81% when the multiplicity of infection is 400 pfu/cell;the cell transfection with recombinant adenovirus carrying wild-type p53,GM-CSF and B7-1 genes can express the gene products;cell proliferation was reduced after transfection with the recombinant adenovirus carrying wild-type p53,GM-CSF and B7-1 gene.Conclusion The adenovirus vector can transduce wild-type p53,GM-CSF and B7-1 genes into ovarian cancer cell line SKOV-3 effectively;wild-type p53,GM-CSF and B7-1 genes modification can significantly reduce the proliferation of SKOV-3 cells.
