高频电刺激吗啡精神依赖大鼠伏隔核后相关脑区细胞外氨基酸类神经递质的变化

Changes of Extracellular Amino Acids Levels in Related Brain Regions after High Frequency Electrical Stimulation of Nucleus Accumbens in Morphine Psychic Dependent Rats

摘要:

目的 通过研究高频电刺激吗啡精神依赖大鼠伏隔核(nucleus accumbens,NAc)后,NAc、苍白球腹侧核(ventral pallidum,VP)以及中脑腹侧被盖区(ventral tegmental area,VTA)中细胞外谷氨酸(glutamate,Glu)和γ氨基丁酸(gamma-aminobutyric acid,GABA)含量的变化,阐述高频电刺激治疗药物精神依赖的机制。方法 Wistar大鼠皮下注射盐酸吗啡10 d(3次/ d),对照组大鼠在同等时间给予同体积的0.9%氯化钠注射液皮下注射。停药后第7天将大鼠麻醉,立体定向引导下将刺激电极置入单侧NAc,在刺激前后用微透析探针对同侧NAc,VP和VTA行微透析,用高效液相色谱法(high performance liquid chromatography,HPLC)测定透析液中的Glu和GABA的含量。对照组大鼠只植入电极,不刺激。结果 吗啡组NAc中Glu含量增高。对NAc行高频电刺激后,NAc和VTA中细胞外Glu含量递减,GABA含量无明显变化;VP中细胞外Glu和GABA含量无明显变化。结论 Glu在维持吗啡精神依赖的过程中发挥重要作用,高频电刺激能通过减少NAc和VTA中细胞外Glu的含量来消除吗啡精神依赖。

Abstract:

Objective To investigate the changes of levels of glutamate(Glu) and gammaaminobutyric acid(GABA) in nucleus accumbens(NAc), ventral pallidum(VP) and ventral tegmental area(VTA) after high frequency electrical stimulation of NAc in morphine psychic dependent rats, and illustrate the mechanism of high frequency electrical stimulation treatment of drug psychic dependence. Methods Morphine hydrochloride was injected subcutaneously in Wistar rats for 10 days(3 times per day), sodium chloride solution was injected subcutaneously in rats of control group. At the 7th day after withdrawal of morphine, Concentric bipolar electrodes were stereotaxically implanted into the NAc and microdialysis probes were unilaterally placed into NAc, VP and VTA of rats after anesthesia, the microdialysates before stimulation and after stimulation were collected. The levels of extracellular Glu and GABA in microdialysates were determined by high-performance liquid chromatography(HPLC). In control group, only stimulating electrodes were implanted, stimulation was not proceeded.Results The levels of extracellular Glu in NAc increased in morphine group. During and after stimulation of NAc, the levels of extracellular Glu in NAc and VTA decreased, but the levels of extracellular GABA did not change; The levels of extracellular Glu and GABA in VP did not change. Conclusion Glu is an important factor to maintain psychic dependence, and high frequency electrical stimulation can induce decrease in extracellular Glu levels in NAc and VTA to diminish morphine psychic dependence.
