大肠杆菌甲酸脱氢酶脱钼前体及其复钼研究

摘要:

大肠杆菌(Escherichia coli)在含钨介质中生长时不具有甲酸脱氨酶活性,但当菌体在蛋白质合成被抑制的条件下,除去钨盐并加钼盐共同保温时,可以检测到甲酸脱氢酶的存在。提示在含钨介质中菌体能合成无活性的甲酸脱氢酶脱钼前体。实验对这种前体进行了纯化和复钼研究,并为研究金属蛋白分子结构与功能关系提供了一种有用的实验材料。本实验仍在继续进行。

Abstract:

The present study proved that Escherichia coli synthesized ademolybd-precursor of Formate dehychogenase (FDH) in culture media containing tungstate under anaerobic condition. The whole cell conld restore FDH activity when tungstate was removed and molybdate was added. But purified demolybdo-enzyme would not restore its activity under he same condition. The experiment provides a useful material for the study of metalion incorporateing into apoenzyme.
