氟伐他汀对高脂血症模型兔血脂的影响及其与内膜增生的关系

Effects of Fluvastatin on Serum Lipid and its Relationship with Intimal Hyperplasia in the Hyperlipidemic Model of Rabbit after Iliac Artery Endothelial Denudation

摘要: 为探讨氟伐他汀对血脂的影响及其与内膜增厚的关系,将髂动脉内膜剥脱术后高脂血症模型兔随机分为氟伐他汀组和非用药组(各组n=28),并设正常对照组(n=7).分别于喂高脂饮食前及动脉剥脱术后1、2、4、8周,观察氟伐他汀组和非用药组动物血清TC、TG及LDLC的变化;并观察动脉剥脱术后1、2、4、8周氟伐他汀组和非用药组兔髂动脉切片的新生内膜、中膜厚度及内膜/中膜比值(I/M).结果:氟伐他汀组血清TC、TG、LDLC浓度明显低于非用药组(P<0.05);氟伐他汀组的新生内膜厚度和I/M明显低于非用药组(P<0.01);动脉内膜厚度与TC、TG、LDLC呈明显的直线相关(偏相关系数分别为0.911、0.892、0.942,P<0.01).提示:氟伐他汀可调节血脂,有效抑制血管内膜过度增生.

Abstract: To investigate the effects of fluvastatin on blood lipid in inhyperlipidemic model of rabbit after iliac artery endothelial denudation and the relationship between serum lipid and intimal thickening in the study, the New Zealand white rabbits were randomized into three groups: drug treatment group(n=28),non drug treatment group(n=28)and control group(n=7). The atherosclerotic lesions were developed by endothelial denudation with balloon catheter and high dosage of cholesterol diet at the same time. The experimental animals were killed and iliac arteries were removed at 1、2、4、8 weeks after endothelial injury respectively and bloodsamples were taken at the same time. By the methods of routine HE staining, blood lipid measure and image analysis system, observed the effects of fluvastatin on the level of serum TC、TG、LDL-C and the thickness of intima and media,intima/media(I/M) ratio and the relationship between serumlipid and intimal thickening. The level of TC、TG、LDL-C in drug treatment group was remarkably lower than that in non drug treatment group(P<0.05). The thickness of neointima and I/M in drug treatment group was much thinner than that in non drug treatmeant group. Coefficient of linear correlation between intimal thickness and TC、TG、LDL-C were 0.911,0.892,0.942 (P< 0.01) respectively.Fluvastain can lower blood lipid apparently, which may relate its effect to inhibiting excessive thickening of vascular intima.
