固本通络汤对糖尿病肾病大鼠肾小管间质 p38MAPK表达的影响

Effects of Gubentongluo decoction on tubulointerstitial p38MAPK expression in diabetic nephropathy rats

摘要: 目的 观察固本通络汤对糖尿病肾病(diabetic nephropathy,DN)大鼠肾小管间质p38丝裂原活化蛋白激酶(p38MAPK)表达的影响。方法 采用单侧肾切除加链脲佐菌素诱导大鼠DN模型,将其48只DN模型大鼠分为正常组、模型组、氯沙坦组和固本通络汤大、中、小治疗组,观察固本通络汤对DN大鼠血糖、肾功能、肾小管病理损伤及肾小管间质p38MAPK表达的影响。结果 固本通络汤对DN大鼠血糖、肾功能、肾小管病理损伤均有较好的改善作用,并能明显降低肾小管间质p38MAPK的表达。结论 固本通络汤能够明显降低肾小管间质p38MAPK表达,延缓DN大鼠肾脏病理进程,对肾脏具有一定的保护作用。

Abstract: Objective To study the effects of Gubentongluo decoction on Tubulointerstitial p38MAPK expression in diabetic nephropathy rats. Methods The model of diabetic nephropathy rats was established by unnephrectomized and intraperitoneal injection of streptozotocin(STZ). The rats were divided into six groups randomly, including normal group, model group, western medicine group, Gubentongluo decoction (high, middle, low dosage) groups. Then, the levels of blood glucose and renal function(including serum creatinine, urea nitrogen) were measured in each group of rats. HE staining was applied to observe changes in tubules. Immunohistochemistry was used to observe tubulointerstitial p38MAPK expression. Results Compared with the model group, intragastric administration of Gubentongluo decoction significantly lowered the blood glucose, serum creatinine, urea nitrogen, improved the tubular pathological changes and reduced tubulointerstitial p38MAPK expression. Conclusion Gubentongluo decoction could significantly reduce tubulointerstitial p38MAPK expression, delay the process of renal pathology in DN rats, and has a protective effect on kidney.
