摘要：

目的观察固本通络汤对糖尿病肾病大鼠（diabetic nephropathy，DN）转化生长因子-β₁ （transforming growth factor β₁，TGF-β₁）、基质金属蛋白酶-9（matrix metalloproteinase-9，MMP-9）、基质金属蛋白酶抑制因子-1（tissue inhibitors of metalloproteinases-1，TIMP-1）表达的影响。方法采用单侧肾切除加链脲佐菌素制备DN模型，分为正常对照组、模型对照组、氯沙坦组和固本通络汤大、中、小治疗组，免疫组织化学法检测大鼠肾脏TGF-β₁、MMP-9、TIMP-1的表达。结果大、中剂量的固本通络汤能明显降低TGF-β₁、TIMP-1的表达（P<0.05），升高MMP-9的表达（P<0.01）。结论固本通络汤可能通过抑制DN大鼠肾组织TGF-β₁的表达，调节MMP-9、TIMP-1的平衡，发挥对DN大鼠的肾脏保护作用。

Abstract:

Objective To study the effects of Gubentongluo decoction on transforming growth factor(TGF-β₁)、matrix metalloproteinase-9 (MMP-9) and tissue inhibitors of metalloproteinases-1(TIMP-1) expression in diabetic nephropathy rats. Methods The model of diabetic nephropathy rats was established by unnephrectomized and intraperitoneal injection of streptozotocin(STZ).The rats were divided into six groups randomly, including normal group, model group,western medicine group, Gubentongluo decoction (high, middle, low dosage) groups. The immunohistochemical method was applied to detect the expression of TGF-β₁、MMP-9 and TIMP-1. Results Compared with the model group, the high and middle groups of Gubentongluo decoction significantly lowered the expression of TGF-β₁ and TIMP-1(P<0.05), increased the expression of MMP-9(P<0.01).Conclusion Gubentongluo decoction could significantly inhibit the expression of TGF-β₁,regulating the balance of MMP-9 and TIMP-9, delaying the process of renal pathology in DN rats,having a protective effect on kidney.
