旋毛虫抗原基因Ts87在毕赤酵母系统中优化表达的研究

Study on the Expression Conditions of Ts87 Antigenic Gene from Trichinella spiralis in Pichia pastoris

摘要: 目的 优化旋毛虫抗原基因Ts87在毕赤酵母系统中的表达,提高其表达量.方法 选择去除自身信号肽序列的Ts87基因构建天然N末端的重组质粒,电击转化经锂盐处理过的酵母细胞X-33,摸索诱导起始细胞浓度、甲醇诱导浓度及其添加的时间间隔、pH值及诱导时间等条件对重组菌株表达的影响,并对表达产物进行Western blotting鉴定.结果 采用锂盐处理毕赤酵母细胞的感受态制备方法可以极大的提高电击转化效率,并确定pPICZaA-Ts87nature/X-33株最佳表达条件为:OD600 nm=1.0,1.0%/24 h体积比添加甲醇,pH值7.0或8.0,诱导表达72 h.将此条件下的培养物上清液30μL直接上样,表达产物可被感染旋毛虫的兔血清所识别.结论 本研究优化了Ts87在毕赤酵母系统中的表达条件,为后续研究工作打下基础.

Abstract: Objective To increase yield expression of Ts87 gene was optimized in P.pastoris from the aspects,such as vector construction,to obtain multicopy transformants and inductive conditions.Methods Signal sequences of Ts87 gene was deleted and the recombinant vector was constructed with nature N end,then used X-33 host cell dealt with LiCl before electroporation to get a appropriate conditions from cell densities,methanol concentration,pH and induction time,etc.The expression product was analyzed by Western blotting.Results The efficiency of electroporation increased by the method which prepared host cell with LiCl and the appropriate inductive conditions were OD600 nm=1.0,1.0%/24 h methanol added,pH=7.0 or 8.0,72 h.The expression protein was detected as positive reaction with infected rabbit sera when tested directly with 30 μL supernatant.Conclusion The appropriate condition for Ts87 gene in P.pastoris expression system is obtained.
