外周血miR-181a与PGRMC1水平与乳腺癌特征相关性研究

Study on the correlation between blood miR-181a and PGRMC1 levels and breast cancer characteristics

摘要:

目的  探讨血清微小RNA-181a(microRNA-181a,miR-181a)与全血孕激素受体膜组分1(progesterone receptor membrane component 1 ,PGRMC1)的表达水平与乳腺癌临床特征的相关性,并评估二者作为乳腺癌风险潜在生物标志物的可能性。方法  募集并收集60例术前乳腺癌患者(病例组)和60例年龄匹配的健康女性(对照组)的血液样本。采用实时荧光聚合酶链式反应(real-time fluorescence polymerase chain reaction,RT-qPCR)和酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测血清miR-181a-5p的相对表达量及全血PGRMC1浓度。分析两组间miR-181a与PGRMC1水平相关性及比较不同临床病理特征亚组间的差异。结果  与健康对照组相比,乳腺癌患者血清miR-181a表达水平显著降低(P<0.001),而PGRMC1浓度显著升高(P<0.001)。在乳腺癌患者中,血清miR-181a水平与PGRMC1浓度呈显著负相关(P<0.01)。组间比较显示,肿瘤直径T3-T4组、临床分期Ⅲ-Ⅳ级、淋巴结转移与肿瘤直径T1-T2、临床分期Ⅰ-Ⅱ、淋巴结无转移相比,血清miR-181a相对表达较低,PGRMC1浓度较高(均P<0.05)。结论  乳腺癌患者血清中miR-181a表达下调,PGRMC1蛋白水平上调,二者呈负相关,血清miR-181a低表达和PGRMC1高表达与肿瘤直径大、组织学分级高、淋巴结转移及临床分期晚等提示肿瘤侵袭性强和预后差的病理特征密切相关,提示联合检测血清miR-181a和PGRMC1可能为乳腺癌的风险预测提供新的思路。

Abstract: ObjectiveTo investigate the correlation between the expression of microRNA-181a (miR-181a) in serum and the progesterone receptor membrane component 1 (PGRMC1) in whole blood, as well as the association with breast cancer characteristics, and to evaluate its potential as a biomarker for breast cancer risk.MethodsBlood samples were collected from 60 pre-operative breast cancer patients and 60 age-matched healthy women. The relative expression level of serum miR-181a and the concentration of PGRMC1 in whole blood were measured using real-time fluorescence polymerase chain reaction (RT-qPCR) and enzyme-linked immunosorbent assay (ELISA), respectively. The correlation between miR-181a and PGRMC1 levels was analyzed in both groups, with further comparisons made among subgroups based on clinicopathological characteristics.ResultsCompared with the healthy control group, the serum miR-181a expression level was significantly decreased in breast cancer patients (P < 0.001), while the PGRMC1 concentration was significantly increased (P < 0.001). Among breast cancer patients, serum miR-181a levels were significantly negatively correlated with PGRMC1 concentrations (P < 0.01). Intergroup comparisons showed that the T3-T4 tumor diameter group, clinical stage Ⅲ-Ⅳ group, and lymph node metastasis group had lower serum miR-181a relative expression and higher PGRMC1 concentrations compared to the T1-T2 tumor diameter group, clinical stage Ⅰ-Ⅱ group, and lymph node non-metastasis group, respectively (all P < 0.05).ConclusionThe expression of miR-181a in the serum of breast cancer patients is downregulated, and the protein level of PGRMC1 is upregulated. They are negatively correlated and closely related to the malignant characteristics of breast cancer, suggesting that the combined detection of serum miR-181a and PGRMC1 may provide a new perspective on risk assessment and prognosis of breast cancer.
