RGDS肽对离体大鼠血小板聚集和ERK1/2磷酸化的影响

摘要:

目的 观察外源性精-甘-天-丝冬氨酸(RGDS)肽对离体大鼠血小板聚集及细胞外信号调节激酶1/2(ERK1/2)磷酸化的影响,探讨RGDS肽对血小板聚集的作用与ERK1/2磷酸化表达改变的关系.方法 用比浊法测定血小板最大聚集率;Western-blot测定血小板ERK1/2磷酸化表达.结果 RGDS肽抑制血小板聚集后伴随ERK1/2磷酸化表达减少.结论 ERK1/2通路参与血小板聚集;抑制ERK1/2磷酸化表达可能是外源性RGDS肽抗血栓的机制之一.

Abstract:

Objective To obsere influence of RGDS peptide on rat platelet aggregation rate and the ERK1/2 phosphorylation induced by different agonists in vivo and study the relationship between the change of ERK1/2 phosphorylation and the change of platelets aggregation with RGDS peptide. Methods Maximal aggregation rate(MAR) was measured with nephelometry; ERK1/2 phosphorylation was detected by Western blot. Results The MAR and phosphorylation of ERK1/2 were inhibited by RGDS peptide. Conclusion The phosphorylation of ERK1/2 was one of the cellular signal transduction mechanisms of the platelets aggregation; and the inhibition of ERK1/2 phosphorylation of may be one of the mechanisms that RGDS peptide antagonize thrombus.
