面神经损伤后热休克蛋白27在面运动核团的表达变化

Change in Expression of Heat Shock Protein 27 in Facial Motor Nucleus after Facial Nerve Axotomy

摘要: 目的 研究切断面神经后热休克蛋白27(heat shock protein 27,HSP27)在面运动核团内表达的变化情况;探讨面神经损伤后面神经功能恢复的分子机制。方法 用随机数字表法将30只Wistar大鼠分为3组:实验组、手术对照组、空白对照组,每组10只;在切断右侧面神经术前和术后的第1、2、3、4天,获取不同组内2只大鼠右侧的面运动核团,从核团组织中提取包含热休克蛋白27在内的上清液,用Western blotting方法测定上述不同实验组的组织上清液内热休克蛋白27的表达。结果 切断面神经术后第1天实验组,大鼠面运动核团内热休克蛋白27表达比切断前有明显提高,术后第2天表达达到高峰,术后第3天表达开始下降,术后第4天表达较术后第3天继续下降,但仍高于术前;手术对照组和空白对照组HSP27的表达无明显变化。结论 切断面神经后,面运动核团内热休克蛋白27表达上调,术后第2天表达达到高峰,之后递减到术后第4天,但表达始终高于术前。

Abstract: Objective To study the change in the level of heat shock protein 27 in facial motor nucleus after facial nerve axotomy. Methods Thirty Wistar rats were randomized into 3 groups: experimental group, surgical control group, and blank control group, with 10 rats in each group; at the time points of pre- and 1, 2, 3, 4 days post-right facial nerve axotomy, the samples of right facial motor nucleus of 2 rats in every group were collected; the supernatant containing heat shock protein 27 from the collected tissue samples were collected and the level of heat shock protein 27 in the three different experimental groups were determined by the use of Western blotting. Results In the experimental group, the expression level of heat shock protein 27 in facial motor nucleus was significantly up-regulated on the 1st day post- facial nerve axotomy as compared with pre- facial nerve axotomy, peaked on the 2nd day post-facial nerve axotomy, declined on the 3rd day post-facial nerve axotomy, and further declined on the 4th day post-FNA, which was still higher than the expression level of pre- facial nerve axotomy; in the surgical and blank control group, there was no significant difference in the level of heat shock protein 27. Conclusion After facial nerve axotomy, the level of heat shock protein 27 was obviously up-regulated in facial motor nucleus, peaked on the 2nd day post-facial nerve axotomy, and then gradually declined on the 3rd and 4th day post-facial nerve axotomy, but still remained higher than the level of pre-facial nerve axotomy.
