标本保存方法和时间对血清总二氧化碳结果的影响

摘要:

目的 研究不同的标本保存方法和时间对血清总二氧化碳(total carbon dioxide,TCO2)检测结果的影响。方法 保证TCO2检测系统稳定性符合实验室质量目标的前提下,利用高值QC(quality control)、低值QC、混合血清3种样本,分别检测室温敞口、室温封口、4℃敞口、4℃封口时不同时间点TCO2的浓度,计算TCO2浓度随时间的变化率。结果 高值QC、低值QC、混合血清在室温敞口保存75 min时,TCO2结果分别下降了9.65%、13.42 %、7.74%;4℃敞口保存75 min时,TCO2结果分别下降了8.96%、8.60%、3.94%;在室温封口保存90 min时,TCO2结果分别下降了4.53%、9.43%、3.94%;在4℃封口保存2 h,TCO2结果仅分别下降了3.44%、3.56 %、2.19%。结论 随着标本保存时间的延长血清TCO2浓度不断下降,并且不同的保存方法下降的速率不同。封口保存TCO2结果下降低于敞口状态,保存温度越低TCO2结果下降越慢。

Abstract:

Objective To study the effect of sample storage method and time on the test results of serum total carbon dioxide. Methods Under the condition that total carbon dioxide(TCO2) measurement system can guarantee the stability of TCO2 results. TCO2 of two quality controls and a mixed serum was measured promptly at different time points after storage under various conditions: open samples at room temperature(25±2)℃, sealed samples at room temperature, open samples at 4℃, and sealed samples at 4℃. Results High level quality control(QC), low level QC and mixed serum samples decreased by 9.65%, 13.42% and 7.74%, respectively, at 75 min when samples were stored at room temperature and kept open. The same samples decreased by 4.53, 9.43 and 3.94% at 90 min when stored at room temperature and kept seal. The samples decreased by 8.96, 8.60 and 3.94% at 75 min when stored at 4℃ and kept open, and decreased by 3.44%, 3.56% and 2.19% at 2 h when stored at 4℃ and kept sealed. Conclusion TCO2 decreased over time and decreased at different rate according to storage condition. TCO2 of sealed samples decreased much slower than open samples. The colder samples stored, the slower TCO2 decreased.
