体外培养成人及成年大鼠肾上腺嗜铬细胞

摘要: 成人及成年大鼠肾上腺髓质组织比较致密,消化困难,应选用消化能力较强的胶原酶。在体外培养条件下,嗜铬细胞较其它类型细胞贴壁能力差,以多聚赖氨酸或鼠尾胶原包被底壁有利于细胞贴壁生长。神经生长因子(nGF)可促进嗜铬细胞贴壁,存活和生长出神经轴突样突起,但是成人及成年大鼠肾上腺嗜铬细胞对于NGF的依赖程度存在种属差异。在合NGF的培养基中大鼠嗜铬细胞存活数量增加,存活时间延长,并有较多细胞发育出神经轴突样突起。成人嗜铬细胞对NGF也产生一定反应,但其依赖程度不如鼠嗜铬细胞明显。

Abstract: The in vitro culture of adult human and rat's chromaffin cell was succeeded by the authors. Collagenase should be used for dissociation, because both tissues were dense. Chromaffin cells showed lower attachment ability then other cells. It may be improved by coating the culture dishes with polysine or collagen. NGF could enhance the attachment, Survival and neurite outgrowth, however, there appeared species differences. The presence of NGF in medium could enhance the survrval of rat chromaffin cells, prolong Survival time, promote neurite outgrowth. The human chromaffin cell responsed to NGF in a certain degree, but the responses were not so obvious as those of the rat.
