成年与胚胎大鼠卵泡细胞的体外研究

摘要:

将胚胎、成年和妊娠Wistar大鼠的卵泡细胞进行冷冻保存、复苏后在RPMI640培养液中培养,观察各自的生长发育情况。形态学显示:冷冻复苏的各组卵泡细胞均有贴壁增殖生长的能力,培养初期胚胎与成年卵泡的贴壁生长速度快,而妊娠卵泡细胞贴壁生长发生的较晚;但培养至4d后,胚胎卵泡细胞贴壁生长能力逐渐减弱并消失。组织化学显示:培养3d后的胚胎卵泡细胞3-β-羟甾脱氢酶(3-β-HSDH)呈阴性,酸性磷酸酶(ACP)和PAS反应也弱于成年卵泡细胞。结果提示:胚胎卵泡细胞可能是未完全成熟,但已具有内分泌潜能的细胞.

Abstract:

Refrigerated follicular cells that were taken from ovaries of embryonal, adult and pregnant rats were resuscitated and cultured in RPMI 640. The morphological study indicated that all the resuscitated cells had the proliferative capability in culture medium. But the embryonal follicular cells lost the proliferative capability from the 4th day in culture. The cytochemical study showed that embryonal follicular cells were negative to 3-β-HSDH from the 3rd day. The study demonstrated that embryonal follicular cells are immature cells, but they have potential endocrine function.
