高密度无血清培养肝细胞的研究

Hepatocytes Cultured in Serum-free Medium at High-density for Bioartificial Liver

摘要: 利用高密度无血清培养肝细胞,以观察体外肝细胞形态和分化特点、白蛋白合成及其mRNA表达,为生物人工肝提供理想的肝细胞来源。结果:培养肝细胞48h后逐渐形成肝板样网络结构,伴胆小管形成。原位杂交显示所培养肝细胞具有良好的白蛋白mRNA表达。结果提示高密度无血清培养的肝细胞可形成类似体内肝细胞的形态和功能,可为生物人工肝提供理想的肝细胞来源。

Abstract: To seek ideal source of hepatocytes for bioartificial liver, we cultured hepatocytes in serum-free medium at high density. The morphological and functional characteristics of hepatocytes were observed. Hepatocytes showed the gradual development of bile canaliculi-like structures into an anastomatic network and high level of albumin mRNA expression by in situ hybridization was observed. The results suggests that hepatocytes cultured in serum-free medium at high-density can reconstruct in vivo-like structure and function and can be used as an ideal source for bioartificial liver.
