细胞因子对K562细胞HSP gp96基因及蛋白表达的影响

摘要: 目的 研究细胞因子与K562细胞共孵育时不同浓度与不同时间条件对gp96基因及蛋白的影响.方法 分别将浓度为0 kU/L、200 kU/L、400 kU/L、600 kU/L、800 kU/L、1 000 kU/L的IFN α及0 μg/L、2 μg/L、4 μg/L、6 μg/L、8 μg/L、10 μg/L的IFN γ与K562细胞共孵育12 h,用Western blot及RT PCR法分析其中的gp96 mRNA和蛋白.分别将浓度为600 kU/L的IFN α和6 μg/L的IFN-γ与K562细胞共孵育0、3、6、9、12、18、24、36 h检测gp96 mRNA含量.结果 1)不同浓度IFN α和IFN γ分别与K562细胞共孵育12 h后,hsp gp96基因及其蛋白的表达出现相似的改变,即随着浓度的增加表达量逐步增加,当浓度达IFN α 600 kU/L、IFN γ 6 μg/L后,表达量逐步下降.2)IFN α 600 kU/L、IFN γ 6 μg/L分别与K562细胞共孵育,细胞中hsp gp96 mRNA表达量均先增加后逐步下降,在9 h达高峰. 结论IFN α、IFN γ与K562细胞共同孵育,能通过上调细胞中hsp gp96 mRNA增加HSP GP96蛋白.

Abstract: Objective Tumor-derived heat shock protein glycoprotein 96(HSP GP96) can be used as vaccines for refractory cancers. The effects of cytokines on the gene and protein expressions of GP96 in K562 cells at different concentrations and different hours were investigated. Methods K562 cells were coincubated with different concentrations of cytokines for 12 hours. GP96 mRNA and GP96 proteins of K562 cells were analyzed. Total RNA of K562 cells, which were respectively coincubated with concentrations increases, but they descend at certain concentrations(IFN-α 600 kU/L or IFN-γ 6 μg/L at different hours, were analyzed. Results 1) Dose-effect: changes of the gene and protein expressions of GP96 from K562 cells which were respectively coincubated with different concentrations of IFN-α and IFN-γ for 12 hours were similar: they gradually increased with the concentrations, but descended at certain concentrations(IFN-α 600 kU/L, IFN-γ 6 μg/L). 2) Time-effect: steady level of GP96 mRNA were achieved up to 9 hours of continuous exposure to IFN-α 600 kU/L and IFN-γ 6 μg/L, and then descended gradually. Conclusion IFN-α and IFN-γ can increase the protein expressions of GP96 from K562 cells by increasing gp96 gene expression.
