过氧化氢和帕金森病患者血清促进α-突触核蛋白向线粒体与细胞核转运

H2O2 and Serum of Patient with Parkinson's Disease Promote Mitochondrial and Nuclear Translocation of Alpha-synuclein

摘要: 目的 研究氧化应激和帕金森病(Parkinson's disease,PD)患者血清对α-突触核蛋白(alpha-synuclein,α-Syn)向线粒体以及细胞核转运的影响。方法 将MES23.5多巴胺能神经细胞在含5%胎牛血清的DMEM/F12培养基中培养,然后分别用过氧化氢(H2O2)以及PD和正常人血清处理,免疫荧光标记法观察不同处理对α-Syn向线粒体以及细胞核的转运以及对线粒体及细胞核的影响。比色法测定血清超氧化物歧化酶(superoxide dismutase,SOD)、脂质氧化(malondialdehyde,MDA)和总抗氧化能力(totalanti-oxidation competence,T-AOC),观察血清的氧化应激水平。结果 H2O2和PD血清促进α-Syn向线粒体转运与细胞核转运,并造成线粒体皱缩、崩解,PD血清处理同时引起类似凋亡的核碎裂现象。PD血清的SOD、MDA和T-AOC高于对照血清。结论 氧化应激和PD血清均可促进α-Syn向线粒体与细胞核转运,PD血清促进α-Syn向线粒体与细胞核转运可能与其氧化应激水平增高有关。

Abstract: Objective To investigate the effects of oxidative stress and serum of patient with Parkinson's disease(PD) on the mitochondrial and nuclear translocation of alpha-synuclein(α-Syn).Methods MES 23.5 dopaminergic neuronal cells were cultured in DMEM/F12 medium containing 5% fetal bovine serum.The cells were treated with H2O2 and sera from PD patients and the mitochondrial and nuclear translocations of α-syn were examined using immunofluorescent labeling.The serumal oxidative stress was evaluated by measuring with colorimetry the levels of superoxide dismutase(SOD),malondialdehyde(MDA),and total anti-oxidation competence(T-AOC) in the serum.Results Both the H2O2 and PD serum promoted the mitochondrial and nuclear translocations of α-syn,with shrank and disrupted mitochondria observed in the treated cells.The PD serum-treated cells also displayed apoptosis-like disintegrated nuclei.The levels of SOD,MDA and T-AOC were higher in PD sera than in normal sera.Conclusion Oxidative stress and PD serum promote mitochondrial and nuclear translocations of α-syn.The effect of PD serum is probably related to the increased oxidative stress.
