牛嗜铬细胞体外培养及其分泌多巴胺的测定

摘要: 为给肾上腺髓质细胞移植治疗帕金森病提供理论依据,观察正常体外培养和冷冻复苏后牛肾上腺嗜铬细胞的形态变化,并检测培养液中的多巴胺含量。结果:悬浮培养嗜铬细胞在培养液中集聚成团,始终保持圆形,并分泌多巴胺,在冷冻复苏后仍存活;单层贴壁培养嗜铬细胞于贴壁后开始变形,伸出短的突起,但于1周后开始退化;体外培养嗜铬细胞分泌多巴胺在5~6d达高峰。结果提示:肾上腺嗜铬细胞可作为脑内移植治疗帕金森病的细胞来源.

Abstract: The morphological changes of cultured bovine chromaffin cells were observed and the dopamine secreted was determined by HPLC to explore the feasibility of gene therapy with adrenal medulla cell transplantation. Results: The chromaffin cells in suspension culture, aggregated in round shape all the time and could secrete dopamine, and were still alive after resuscitation from cryopreservation; the adhesion cells showed round shape first, then stretched out short processes, turned in to shapes as triangle, fusiform, and polygon et al and restored to round shape after a week of culture. The HPLC results showed that the dopamine content arrived at the peak at about 5~6 days of culture. Adrenal chromaffin cell is a valuable source that can be used for gene therapy.
