凝血酶对大鼠离体胸主动脉和肺动脉的收缩作用及其可能机制

摘要:

目的 观察凝血酶对离体大鼠胸主动脉与肺动脉血管张力的影响,并初步探讨其可能的作用机制。
方法 采用Powerlab生物信号采集放大系统记录离体大鼠肺动脉与胸主动脉的张力信号,利用RT-PCR技术检测血管壁凝血酶受体的表达。
结果 凝血酶对离体大鼠肺动脉及胸主动脉具有明显的收缩作用,且该收缩作用与凝血酶受体-1激动剂的作用相似,不依赖于细胞外钙离子的存在。大鼠肺动脉及胸主动脉血管壁中均有凝血酶受体-1的表达。
结论 凝血酶对大鼠离体血管具有收缩作用并且可能是通过凝血酶受体-1而起作用,该作用不依赖于细胞外钙离子的存在。

Abstract:

Objective To explore the vasoconstrictive effect and mechanism of thrombin on pulmonary artery and thoracic aorta of rats.
Methods The pulmonary artery and thoracic aorta ring of rat were isolated in vitro according to the procedures routinely used. Vascular constriction tension recordings were accomplished by using Powerlab biology signal collection system. RT-PCR method was used to detect the mRNA expression of thrombin receptor in vascular rings.
Results The pulmonary artery and thoracic aorta rings were constricted by thrombin which was independent on the extracellular calcium. This effect was similar to that of the TRAP6, a thrombin receptor agonist. The mRNA expression of thrombin receptor-1 can be detected in the pulmonary artery and thoracic aorta.
Conclusion Thrombin induced pulmonary artery and thoracic aorta ring constriction was independent of extracellular calcium and might be mediated by thrombin receptor 1(PAR1).
