高效液相色谱-质谱联用分析法测定血清全氟辛烷磺酸

Study on Method for Determination of Perfluorooctane Sulfonate Concentration in Serum Using High Performance Liquid Chromatography-Electrospray Tandem Mass Spectrometry

摘要: 目的 全氟辛烷磺酸(perfluorooctane sulfonate,PFOS)是全氟表面活性剂,被广泛用于纺织、皮革、涂料、农药等生产中,具有较高的生物蓄积性和多种毒性,是目前最难降解的持久性环境有机污染物之一,由其造成的污染已经逐渐成为全球性的环境问题。建立高效液相色谱-质谱联用分析方法对于检测环境和生物体内的PFOS具有实际意义。方法 样品经固相萃取、液相色谱分离后进行电喷雾质谱分析,在多反应监控(multiple reaction monitoring,MRM)模式下使用内标法测定PFOS。结果 PFOS在质量浓度为0 μg/L~120 μg/L时有良好的线性关系,线性相关系数r=0.991 039。由标准曲线计算PFOS的回收率为76.88%。结论 用高效液相色谱-质谱联用内标法定量分析血清中PFOS的浓度灵敏度高,分析速度快,适于血清PFOS含量的测定。

Abstract: Objective To develop an analytical method for the determination of perfluorooctane sulfonate(PFOS). Methods In the procedure a solid-phase extraction for the isolation of PFOS was employed. PFOS was subsequently analyzed by high performance liquid chromatography coupled with electrospray tandem mass spectrometry internal standard method. Mass spectral acquisition was applied with multiple reaction monitoring(MRM). Results The calibration curve was linear over a range from 0 μg/L to 120 μg/L with a correlation coefficient of 0.991 039. The average recovery for PFOS was 76.88%. Conclusion The method enables the precise determination of standards and can be applied to detect PFOS in human plasma samples for monitoring human exposure.
