miR-29s通过下调大麻素受体1抑制ACEA促J774A.1细胞迁移的活性

miR-29s inhibit ACEA-induced J774A.1 cell migration by down regulation CB1 expression

摘要: 目的 研究miR-29a-3p、miR-29b-3p和miR-29c-3p是否可以影响J774A.1细胞中大麻素受体1(cannabinoid receptor type 1,CB1)的表达以及其迁移功能。方法 RT-qPCR法检测CB1 mRNA表达;Boyden chamber法检测J774A.1细胞迁移能力的变化。结果 本研究证明了在J774A.1细胞里,CB1的激动剂ACEA(Arachidonyl-2'-chloroethylamide)可以上调CB1 mRNA的表达,而分别转染了miR-29a-3p、miR-29b-3p和miR-29c-3p的mimics后可以抑制这一过程;同时对J774A.1细胞转染miR-29a-3p、miR-29b-3p和miR-29c-3p的mimics可以阻碍ACEA诱导的J774A.1细胞的迁移。结论 miR-29s通过下调CB1的表达抑制了ACEA促J774A.1细胞迁移的活性。

Abstract: Objective To investigate whether miR-29a-3p, miR-29b-3p and miR-29c-3p affect cannabinoid receptor 1(CB1)expression and cell migration in J774A.1 cells. Methods RT-qPCR was used to detect CB1 mRNA expression in J774A.1 cells. Boyden chamber assay was used for the detection of cell migration. Results In this study, we presented evidences that CB1 agonist Arachidonyl-2'-chloroethylamide (ACEA) upregulates the expression of CB1 mRNA in J774A.1 cells. Whereas miR-29a-3p or miR-29b-3p or miR-29c-3p mimics blocks the increase of CB1 mRNA level and ACEA-induced cell migration in J774A.1 cells. Conclusion miR-29s inhibit ACEA-induced J774A.1 cell migration via downregulating CB1 expression.
