miR-26a在Y79视网膜母细胞瘤细胞系内下调抑癌基因Rb1表达

Tumor suppressor gene Rb1 was down-regulated by miR-26a in Y79 retinoblastoma cells

摘要:

目的 探讨视网膜母细胞瘤细胞系内抑癌基因Rb1表达下调的分子机制。方法 本研究利用real-time PCR和Western blotting等方法分别检测miR-26a和Rb1基因的表达。结果 本研究结果显示在人视网膜母细胞瘤组织细胞与正常人视网膜血管内皮细胞间比较,miR-26a表达差异有统计学意义(P<0.005)。miR-26a可以通过与Rb1基因的3' UTR作用而导致人视网膜母细胞瘤细胞内Rb1基因表达下降。结论 在Y79 Rb 细胞内miR-26a下调抑癌基因Rb1的表达。

Abstract:

Objective To explore the mechanism on down-regulation ofRb1expression in Rb cells. Methods In our study, the expression of miR-26a andRb1was detected by real-time PCR and Western blotting, respectively. Results Here, it was shown that miR-26a was differentially expressed in human retinoblastoma cells and tissues, compared with human retinal microvascular endothelial cells. Our results also showed that miR-26a tightly regulated the expression ofRb1by specifically targeting a conserved sequence motif in its 3' UTR, resulting in a low expression ofRb1in retinoblastoma cells. Conclusion The expression of tumor supressor geneRb1was down-regulated by miR-26a in Y79 Rb cells.
