Establishment of hepatic metastasis model of colorectal cancer for optical in vivo imaging in mice with bioluminescence and near-infrared luminescence

摘要:

目的 利用表达荧光素酶基因的稳定细胞株及小动物活体成像平台,观察小鼠SL4结肠癌肝转移及血管新生情况。方法 利用表达荧光素酶SL4-luc结肠癌细胞株,通过小动物活体成像系统检测结肠癌肝转移的情况,通过血管新生的近红外线探针观察肝转移灶中血管新生情况。结果 利用小动物活体成像系统,生物发光技术确定了结肠癌细胞肝转移,近红外线探针活性确定了肝转移的灶中血管新生。结论 在结肠癌肝转移过程中,应用小动物活体成像技术,确定了结肠癌的肝转移及血管新生。

Abstract:

Objective To detect hepatic metastasis and tumor angiogenesis in vivo imaging with bioluminescence and fluorescence in hepatic metastasis model of colorectal cancer. Methods Mice were inoculated with SL4 cells, colorectal cancer cell line, stably expressed firefly luciferase gene, after 2 weeks of the injection we detect hepatic metastases in vivo imaging in mice with bioluminescence and also determine tumor angiogenesis with in vivo imaging in mice with fluorescence. Results We confirmed hepatic metastasis over time with luciferase activity, and also found tumor angiogenesis in hepatic metastasis foci, which confirmed by immunostaining of CD34 marker of vessels.Conclusion Establishment of hepatic metastasis model of colorectal cancer for optical in vivo imaging in mice with bioluminescence and fluorescence.
