血管紧张素Ⅱ对树突状细胞功能的影响

Ang Ⅱ activates dendritic cell to produce immune function

摘要: 目的 以肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)为对照,探讨血管紧张素Ⅱ(angiotensin Ⅱ,AngⅡ)对树突状细胞(dendritic cells,DC)的功能影响并比较DC对上述刺激的反应差异。方法 体外培养DC2.4细胞,分别加入20 ng/mL TNF-α和100 nmol Ang Ⅱ诱导DC细胞成熟。用MTT、流式细胞仪、Transwell、混合淋巴细胞反应、FITC-Dxtra内吞实验、酶联免疫吸附试验等方法检测上述细胞的增生、分化成熟、迁徙、刺激淋巴细胞增生、内吞和分泌细胞因子白细胞介素-6(interleukin-6,IL-6)、干扰素-γ(interferon-γ,IFN-γ)等方面能力。结果 与TNF-α相比,AngⅡ可以同等程度抑制DC的增生;可以诱导DC成熟标志表达,其能力弱于TNF-α;可以促进DC的迁徙,能力略弱于TNF-α;可以抑制树突状细胞的吞噬能力,其能力弱于TNF-α;促进相关细胞因子IL-6、IFN-γ分泌,与TNF-α作用基本一致;刺激淋巴细胞增生能力增强,其能力弱于TNF-α。结论 Ang Ⅱ具有和TNF-α相近的能力,对于树突状细胞的功能起着正向调节的作用。

Abstract: Objective To investigate the immune effect of angiotensin Ⅱ(Ang Ⅱ) on immature dendritic cells (DC) compared with tumor necrosis factor-α (TNF-α)in vitro. Methods Prepared DC2.4 stimulated with different stimuli, such as 20 ng/mL TNF-α and 100 nmol Ang Ⅱ for indicated time. The effects of TNF-α and Ang Ⅱ on the proliferation, maturation, phagocytosis, cytokine secretion, migration, and communication with T cells of DCs are test by MTT, flow cytometry, ELISA, transwell assay and mixed lymphocyte culture respectively. Results First, the data showed that Ang Ⅱ stimulation significantly inhibited proliferation of DC, induced phenotypic maturation of DC, markedly increased the migration of DC, inhibited macrophage phagocytosis, promoted the secretion of interleukin-6 (IL-6) and interferon-γ(IFN-γ) and initiated the proliferation and activation of T-cell. Conclusion Compared with TNF-α, Ang Ⅱ positively modulated the function of DC to the same extent.
