储存条件对尿触珠蛋白检测的影响

Effects of storage condition on measurement of haptoglobin in urine

摘要: 目的 研究不同标本处理方法及储存条件对尿触珠蛋白(haptoglobin,Hp)检测的影响。方法 收集40例2型糖尿病患者的尿标本,分别进行离心及不离心处理后于室温或4℃放置3、6和24 h,采用ELISA法检测尿Hp浓度。结果 未经离心处理的标本,室温放置3 h和6 h尿Hp浓度较基础值无明显变化(P均>0.05),而24 h尿Hp浓度显著下降(P < 0.01),4℃储存放置3 h较基础值无明显变化(P>0.05),但6 h尿Hp浓度明显下降(P < 0.01)。离心标本在室温和4℃放置3 h稳定(P均>0.05),放置6 h尿Hp浓度明显下降(P均< 0.01)。结论 室温放置6 h的未离心标本可保持尿Hp浓度稳定。冷藏及离心可导致尿Hp浓度的降低。

Abstract: Objective To investigate the effects of different treatments of urinary specimen and storage conditions on urinary haptoglobin levels. Methods The urinary samples of 40 patients with type 2 diabetes were stored at room temperature and 4℃ for 3 hours, 6 hours and 24 hours with and without centrifugation, and then the levels of urinary haptoglobin were measured with enzyme-linked immunosorbent assay. Results For the samples without centrifugation, no change in urinary haptoglobin concentration was observed after 3 hours and 6 hours of room temperature storage(both P>0.05), but there was a significantly decrease after 24 hours(P < 0.01). There was no statistically significant difference in the levels of urinary haptoglobin after 3 hours at 4℃ (P>0.05), and there was a significantly decrease after 6 hours(P < 0.01). The centrifuged samples were stable for 3 hours at both room temperature and 4℃(both P>0.05), but there was a significantly decrease after 6 hours(both P < 0.01).Conclusion It was recommended to store urine samples at room temperature when the measurement of urinary haptoglobin could be completed within 6 hours. Cold storage and centrifugation resulted in the decrease of urinary haptoglobin level.
