Id4对K562细胞增生的影响及其机制的探讨

Effect of Id4 on K562 Cell Proliferation

摘要: 为了探讨Id4基因的反义核酸对人红白血病K562细胞增生的作用,用DNA重组技术将人反义Id4基因插入到真核表达载体pXJ41-neo上,重组质粒和空载体分别转染K562细胞,得到pAId4和pK562(对照)克隆细胞株。发现:pAId4的生长受到抑制,倍增时间加长。免疫印迹实验分析相关蛋白的表达,其中CDK4和CyclinD1在pAId4细胞中的表达量均低于对照细胞pK562,而RB蛋白在pAId4细胞中的表达量高于对照细胞pK562,揭示了反义Id4引起细胞出现生长抑制的可能机制,并探讨了Id4基因对人红白血病K562细胞增生的具体作用。研究结果对造血干细胞的体外扩增、分化及红白血病的基因治疗均有重要的参考价值。

Abstract: In order to study the actions of Id4 in cell proliferation of blood cell, antisense Id4 gene was transfected into human erythroleukemia cell line K562, Id4 gene.Anti-sense Id4 brought about poor growth traits, long multiplication time in comparison with the control group. By using of western-blot, the related protein CDK4, CyclinD1 and RB expressions changed obviously. The results show that Id4 plays an important role in cell proliferation of blood cells.
