Cocktail探针药物评价姜黄对肝细胞色素P450酶的影响

Simultaneous Evaluation of Effect of Rhizoma Curcumae Longae on Hepatic CYP450 by Cocktail Probe Drugs

摘要: 用Cocktail探针药物法,研究姜黄对大鼠肝细胞P450(CYP450)的影响.将SD雄性大鼠分组,给予姜黄的水提物,以生理盐水组为空白对照,以苯巴比妥组为阳性对照,通过HPLC检测Cocktail探针药物的代谢率来评价各组的CYP450.结果:给予姜黄组的大鼠CYP1A2和CYP2E1的水平显著低于对照组,CYP3A4的水平没有显著变化.表明:姜黄对大鼠CYP1A2和CYP2E1有抑制作用,但对大鼠CYP3A4的影响不显著.

Abstract: The aim was to study the change of the rat hepatic CYP 450 content following administration of Rhizoma Curcumae Longae by cocktail probe drugs. Rats were randomly divided into groups. One group of rats received Rhizoma Curcumae Longae plant extracts once daily orally for six days. The rats in another group received once daily orally normal saline as blank control group and phenobarbital as positive control group at the same time. The plasma concentration of cocktail probe drugs was determined by HPLC. The CYP1A2 and CYP2E1 content of the groups of rats received Rhizoma Curcumae Longae plant extracts was reduced, but the CYP3A4 content didn't markedly change. It suggests that Rhizoma Curcumae Longae is inhibitor of the CYP1A2 and CYP2E1, but the CYP3A4 can be hardly affected.
