二价锰对不同月龄大鼠线粒体膜电位影响的体外研究

Effect of Manganese on Mitochondria Membrane Potential of Different-aged Rats in Vitro

摘要: 研究二价锰离子对不同月龄大鼠线粒体膜电位的影响.将不同月龄(4个月和18个月)Wistar大鼠,提取脑线粒体后分为5组(50、100、200、400 μmol/L和空白对照)体外染毒,用荧光光度法连续观察3 min内样品荧光值的变化.结果显示,各染毒组和对照组间差异均有统计学意义(P<0.05);不同月龄大鼠各染毒组间差异也有显著性(P<0.05).随着大鼠年龄和染毒剂量的增加,线粒体膜功能衰退,耐受性降低.

Abstract: The aim was to study the effect of Mn 2+ exposure on membrane function of different month rats brain mitochondrial. Different-aged (4 months and 18 months) Wistar rats's mitochondria were extracted, exposed to MnCl2·4H2O and then divided into five groups(50, 100, 200, 400 μmol/L and the control group). The fluorescence intensity was detected in successive 3 min by fluorophotometric method. Compared with the control group' the exposed groups showed obvious reduction(P<0.05). In addtion, there was significant difference among the drug-receiving groups(P<0.05). Function and resistance of mitochondrial membrane are decreased with the increase of dosage andages.
