香烟烟雾及银杏叶对哮喘大鼠气道TGF-β1表达的影响

Effects of Cigarette Smoke and Ginkgo on the Expression of Transform Growth Factor-β1 in Asthmatic Rats

摘要: 目的 探讨香烟烟雾(cigarette smoke,CS)及银杏叶对哮喘大鼠气道组织转化生长因子-β1(TGF-β1)mRNA和蛋白表达的影响.方法 30只Wistar大鼠随机分为5组:1)哮喘烟熏组(A组);2)哮喘组(B组);3)哮喘烟熏+银杏叶治疗组(C组);4)哮喘+银杏叶治疗组(D组);5)对照组(E组).用蛋白印迹法(Western blot)及反转录-聚合酶链反应(RT-PCR)检测各组气道组织中TGF-β1蛋白和mRNA表达的变化.结果 香烟烟雾可显著增加哮喘大鼠气道组织TGF-β1 mRNA和蛋白的表达,银杏叶则可显著抑制其表达.结论 香烟烟雾使哮喘大鼠气道组织TGF-β1的表达上调,提示银杏叶可能通过降低TGF-β1的表达发挥抑制哮喘的作用.

Abstract: Objective To investigate the influence of cigarette smoke and ginkgo to the expression of transform growth factor-β1 in asthmatic rats.Methods 30 Wistar rats were divided into an asthmatic+CS group(group A),an asthmatic group(group B),an asthmatic+CS+ginkgo group(group C),an asthmatic+ginkgo group(group D) and a control group(group E).The expression of TGF-β1 of bronchial tissue in each group was observed by Western blot and reverse transcription-polymerase chain reaction(RT-PCR),respectively.Results The expression of TGF-β1 from group A and B were significantly increased compared with those of group C,D and E correspondingly and respectively(P<0.01).The expression of TGF-β1 from group D was not increased compared with those of group E(P>0.05).Conclusion CSE can increase the expression of TGF-β1 of bronchial tissue whereas ginkgo can decrease the expression of TGF-β1 of bronchial tissue in asthmatic rats.
