七叶皂苷钠和甲基泼尼松龙对油酸制备急性肺损伤大鼠模型的干预对比研究

A Comparative Study on the Effects of Sodium Aescinate and Medrol on Acute Lung Injury Induced by Oleic Acid in Rat Model

摘要:

目的 比较七叶皂苷钠和甲基泼尼松龙对油酸(oleic acid,OA)制备急性肺损伤(acute lung injury, ALI)大鼠模型的干预作用。
方法 雄性SD大鼠90只,按随机数字表法将其分为9组,分别为空白对照2 h、6 h、18 h组,七叶皂苷钠2 h、6 h、18 h组和甲基泼尼松龙2 h、6 h、18 h组,每组各10只大鼠。采用鼠尾静脉缓慢注射OA(0.1 mL/kg)复制大鼠ALI模型,静推油酸30 min后对七叶皂苷钠组和甲基泼尼松龙组大鼠分别静推七叶皂苷钠2 mg·kg-1和甲基泼尼松龙5 mg·kg-1,空白对照组大鼠尾静脉注射0.1 mL·kg-1 0.9%氯化钠注射液,各组大鼠均在相应处理时间2 h、6 h和18 h后观察。检测指标为动脉血氧分压(arterial partial pressure of oxygen,PaO2)、肺湿/干质量比(wet/dry weight ratio,W/D)、血浆和肺组织匀浆超氧化物歧化酶(super oxide dismutase,SOD)含量、丙二醛(malondialdehyde,MDA)含量、基质金属蛋白酶明胶酶B(matrix metalloproteinases,MMP-9)含量和金属蛋白酶组织抑制因子(tissue inhibitor of metalloproteinase,TIMP-1)含量。
结果 与空白对照组比较,2 h、6 h和18 h时,七叶皂苷钠和甲基泼尼松龙组大鼠PaO2、W/D、SOD、MDA、MMP-9和TIMP-1均无明显变化(P>0.05)。与甲基泼尼松龙组比较,2 h和6 h时,七叶皂苷钠组大鼠PaO2明显升高(P<0.01),18 h时有升高趋势(P>0.05);6 h时,七叶皂苷钠组W/D明显降低(P<0.01),2 h和18 h无明显变化(P>0.05);与甲基泼尼松龙组比较,2 h、6 h和18 h时,七叶皂苷钠组大鼠血浆和肺组织SOD、MDA、MMP-9和TIMP-1均无明显变化(P>0.05)。
结论 七叶皂苷钠和甲基泼尼松龙均对OA制备的急性肺损伤大鼠有抗炎、抗渗出作用;在对ALI大鼠用药初期,七叶皂苷钠作用效果更好,中后期两者作用相当。

Abstract:

Objective To compare the effects of sodium aescinate and medrol on acute lung injury(ALI) induced by oleic acid(OA) in rat model.
Methods Ninety male SD rats were randomly divided into nine groups, which were normal control 2 h, 6 h, 18 h groups, aescinate 2 h, 6 h, 18 h groups and medrol 2 h, 6 h, 18 h groups with 10 rats in each group. ALI models of rats were made by injecting OA(0.1 mL·kg-1) through caudal veins, 30 min later the rats were intravenously injected with aescin sodium(2 mg·kg-1) and medrol (5 mg·kg-1) in two groups. Rats in normal control groups were injected with sodium chloride(0.1 mL·kg-1) through caudal veins. Rats in each group were observed and sacrificed to detect correlated indices at 2 h, 6 h and 18 h after administration. The observed indicators included the partial arterial blood oxygen pressure(PaO2), lung wet/dry weight ratio(W/D), the levels of plasma and lung tissue superoxide dismutase(SOD), malondialdehyde(MDA), matrix metallo-proteinase gelatinase B(MMP-9) and tissue inhibitor of metalloproteinase(TIMP-1).
Results Compared with normal control group, as 2 h, 6 h and 18 h, there were no significant change(P>0.05) in PaO2, W/D, SOD, MDA, MMP-9 and TIMP-1 in aescinate and medrol groups. Compared with medrol group, as 2 h and 6h, PaO2 of rats in aescinate group increased significantly(P<0.01) while there was an increasing tendency as 18 h(P>0.05); As6h, W/D of rats in sodium group decreased significantly(P<0.01) while there were no significant change(P>0.05) at 2 h and 18 h.
Compared with medrol group, at 2 h, 6 h and 18 h, there were no significant change(P>0.05) in plasma and lung tissue SOD, MDA, MMP-9 and TIMP-1 in aescin group.
Conclusion Both aescin and medrol had anti-inflammatory and anti-exudative effect on ALI model of rats induced by OA. In the initial stage of the treatment, aescin had better effect while both had considerable effects in midanaphase.
