雷公藤内酯酮对小鼠变应性接触性皮炎的抑制作用及其机制探讨

Immunosuppressive Effects of Triptonide on Allergic Contact Dermatitis in Mice and the Study of its Mechanism

摘要: 研究了雷公藤内酯酮对小鼠变应性接触性皮大、T和B淋巴细胞增殖反应及脾细胞分泌白细胞介素2(IL-2)活性的影响。发现雷公藤内酯酮1、2、5mg/kg腹腔注射(ip,qd×5d)能显着抑制二硝基氟苯(DNFB)诱导的变应性接触性皮炎,而对脾细胞IL-2分泌活性无显着影响。体外试验表明,0.02、0.10、0.50mg/L浓度的雷公藤内酯酮能显着抑制刀豆素A(ConA)及脂多糖(LPS)诱导的T、B淋巴细胞增殖反应。

Abstract: Triptonide (Tri)extracted from Tripterygium wilfordii Hook inhibited the proliferation of mouse splenocytes induced by suboptimal concentration of concanavalin A or Lipoplysaccharide at concentrations of 0.02, 0.10,and 0.50 mg/L.Although allergic contact dermatitis to dinitrofluorobenzene (DNFB) was inhibited by Tri 1.0, 2.0,and 5.0 mg/kg(ip,qd×5d),the splenocyte interleukin-2 secretion activity of these mouse was not suppressed.The data indicates that Tri has suppressive effects on mouse immune functions and may be used as a potent immunosppressive agents.
