摘要: 为建立一种不需提取线粒体DNA而直接用线粒体进行聚合酶链反应(PCR)的方法,将得到的线粒体用高温加热,使线粒体膜破裂,释放DNA后,直接用于PCR扩增.结果:扩增产物与提纯的线粒体DNA扩增得到的产物一样丰富.提示:该方法适用于对线粒体DNA纯度没有特别要求的试验.

Abstract: The aim was to establish a method for polymerase chain reaction (PCR) of mitochondria without mitochondrial DNA extraction.In high temperature, the membrane of mitochondrial was broken and intracellular DNA leaked out.DNA was analyzed by the PCR method.The amplified product was as much as the product which acquired from purified mitochondrial DNA.The studies suggest that this method is adapted to the common mitochondrial DNA amplified test.
