L-精氨酸对兔气管上皮细胞纤毛运动的影响

Effects of L-arginine on Ciliary Motility in Rabbit Tracheal Epithelial Cells

摘要:

目的 观察一氧化氮(nitric oxide,NO)前体L-精氨酸(L-arginine,L-arg)对兔气管上皮细胞纤毛摆动的影响。方法 体外培养兔气管上皮组织块,采用高速数字化显微成像技术对纤毛摆动进行定量分析,观察10 mmol/L L-arg及100 μmol/L一氧化氮合酶(nitric oxide synthase,NOS)抑制剂N-硝基-L-精氨酸甲酯(N-nitro-L-arginine methyl ester,L-NAME)对上皮纤毛摆动的影响。结果 1 Hanks平衡盐溶液(Hank's balanced salt solution,HBSS)对照组兔气管上皮细胞纤毛摆动在25 min内无明显变化;2 10 mmol/L L-arg处理组纤毛摆动频率(ciliary beating frequency,CBF)随时间延长逐渐增加,其中加药20 min及25 min后,CBF与加药前相比差异有统计学意义;3 100 μmol/L L-NAME预孵育10 min后再加入10 mmol/L L-arg共同孵育25 min,CBF在各个时间点与对照组相比无明显变化;4 100 μmol/L L-NAME单独作用35 min内CBF无明显变化。结论 L-arg可促进兔气管上皮细胞纤毛摆动,其作用机制是通过激活NOS产生NO而实现的。

Abstract:

Objective To investigate the effects of L-arginine on ciliary beat frequency(CBF) in rabbit tracheal epithelial cells. Methods Cultured rabbit tracheal explants were treated with 10 mmol/L L-arginine and 100 μmol/L N-nitro-L-arginine methyl ester(L-NAME), the CBF was measured by high-speed digital microscopy. Results 1 CBF of epithelial cells in Hank's balanced salt solution(HBSS) control group showed no significant changes in 25 min. 2 CBF increased with time in 10 mmol/L L-arginine-treated group, significant changes were observed at 20 min and 25 min treatment. 3 Pretreatment with 100 μmol/L L-NAME for 10 min blocked the increase of CBF by 10 mmol/L L-arginine, CBF showed no significant changes compared with control group. 4 100 μmol/L L-NAME treatment for 35 min did not change CBF of ciliary cells. Conclusion L-arginine increased CBF of rabbit tracheal epithelial cells via the nitric oxide synthase-nitric oxide(NOS-NO) signal pathway.
