不同透析膜对单个核细胞凋亡的影响

Influence of Hemodialyzer Membranes on Apoptosis of Peripheral Blood Mononuclear Cell

摘要: 为了解不同生物相容性透析膜对外周血单个核细胞(PBMC)发生凋亡的影响,分离正常人PBMC,分别与铜仿膜(CU)、gambrane(聚碳酸酯聚乙烯乙二醇共聚物,G)膜孵育,对照组不接触透析膜.在培养的不同时间(12、24、48、72h)收集细胞,应用流式细胞术观察凋亡发生的情况.结果:在细胞孵育48h后,各组PBMC发生凋亡的细胞明显增加,特别是铜仿膜组达(36.31±1.40)%,显着高于另外2组(P<0.05).提示生物相容性差的铜仿膜更易诱导PBMC发生凋亡.

Abstract: This study investigated the influence of hemodialyzer membranes on apoptosis of peripheral blood mononuclear cell. PBMC were seperated from whole blood donated by healthy volunteers . They were incubated in vitro in the presence of either CU membrane or Gambrane membrane . Control cells were incubated in absence of HD membranes. The cells were collected at regular intervals. PBMC apoptosis were evaluated with flowing cetometric analysis. The results indicated that the apoptosis of PBMC in each group increased more obviously than that before the incubation 48 h ago. Apoptosis of CU group was(36.31±1.40)%, significantly higher than other two groups(P<0.05). CU membrane, a bioincompatible HD membrane, can induce the isolated PBMC to apoptosis more easily than others.
