人CD14基因克隆及基因结构分析

Gene Clone and Bio-information Analysis of Human CD14

摘要:

目的 扩增人CD14基因序列,对克隆基因进行结构及生物信息学分析.方法 利用反转录PCR技术从U937细胞总RNA中,扩增编码人CD14的基因序列,对基因结构及其编码的CD14成熟蛋白质进行了限制性酶切位点、蛋白质组成、相对分子质量及等电点、蛋白质稳定性、信号肽定位、蛋白质Prosite和2级结构等生物信息学分析.结果 序列分析结果表明扩增的基因序列与文献报道完全一致,在此基础上获得了人CD14基因结构和生物信息学参数.结论 成功获得人CD14基因综合参数,为进一步重组CD14表达、鉴定和结构功能研究提供了理论依据.

Abstract:

Objective To amplify the gene sequence of CD14, analyze the structure and bio-information of the cloned genes. Methods Human soluble CD14(sCD14) gene cDNA fragment was amplified from total RNA extracted from U937 cells by RT-PCR. The bio-information of cloned CD14 gene and translated protein was then analyzed, DNA and amino sequence, protein prosite and stabilization, alpha-helix/Deleage and Roux were identified. Results The amplified nucleotide sequence fragment consistent completely with the published human CD14 sequence. Moreover, we knew clearly gene structure and bio-information parameter of human soluble CD14 according the sequence. Conclusion The co-index of the gene of CD14 is achieved successfully. These results make it possible to identify Human recombinant CD14 gene.
