胶质瘤患者血浆及肿瘤组织IGFBP-2表达及临床意义

Clinical Significance of Insulin-like Growth Factor Binding Protein-2 in Plasma and Tumor Tissues of Patients with Glioma

摘要: 目的 探讨胶质瘤患者血浆及肿瘤组织IGFBP-2表达及临床意义.方法 应用ELISA法检测92例胶质瘤患者(38例Ⅱ级、24例Ⅲ级和30例Ⅳ级)及18例健康者(对照组)血浆IGFBP-2表达情况;应用单克隆抗体和免疫组化技术,检测上述受试者组织IGFBP 2表达情况.结果 血浆IGFBP-2表达Ⅲ级明显高于Ⅱ级(P<0.001)和正常人(P<0.001),Ⅳ级明显高于Ⅱ级(P<0.001)和正常人(P=0.001),Ⅲ级与Ⅳ级(P=0.562)、Ⅱ级与正常人(P=0.821)之间差异无统计学意义.肿瘤组织中IGFBP-2表达Ⅳ级明显高于Ⅲ级(P=0.006)和Ⅱ级(P<0.001),Ⅲ级明显高于Ⅱ级(P=0.022).肿瘤组织IGFBP-2阳性表达的患者血浆IGFBP-2水平明显高于组织IGFBP-2阴性表达的患者(P=0.017)和正常人(P=0.003),而后两者血浆IGFBP-2水平差异无统计学意义(P=0.262).结论 胶质瘤患者血浆和肿瘤组织IGFBP-2表达可作为胶质瘤患者预后判断的标志物.

Abstract: Objective To investigate the expression of IGFBP-2 in the plasma and tumor tissues of patients with glioma and to evaluate its clinical significance. Methods The expression of plasma IGFBP-2 was tested by ELISA in 92 cases of glioma(grade Ⅱ 38, grade Ⅲ 24 and grade Ⅳ 30) and 18 cases of healthy controls. For the same patients the expression of IGFBP-2 in the paired glioma was investigated by immunohistochemical technique using monoclonal antibody. Results Plasma IGFBP-2 expression in grade Ⅲ was significantly higher than that in grade Ⅱ(P<0.001) and controls(P<0.001), and that in grade Ⅳ was significantly higher than that in grade Ⅱ(P<0.001) and controls(P=0.001). There was no significant difference between grade Ⅲ and grade Ⅳ(P=0.562), and between grade Ⅱ and controls(P=0.821). Tumor IGFBP-2 expression in grade Ⅳ was significantly higher than that in grade Ⅲ(P=0.006) and grade Ⅱ(P<0.001), and that in grade Ⅲ was significantly higher than that in grade Ⅱ(P=0.022). Plasma IGFBP-2 expression in IGFBP-2(+) glioma was higher than that in IGFBP-2(-) glioma(P=0.017) and controls(P=0.003), and no difference was noted between IGFBP-2(-) glioma and controls(P=0.262). Conclusion The expression of IGFBP-2 in plasma and glioma may be a marker for the prognosis of glioma.
