端粒酶反义基因对恶性胶质瘤细胞生长的作用

Effect of Telomerase Antisense Oligodeoxynucleotides on the Growth of Human Malignant Glioma Cells

摘要: 目的 研究端粒酶RNA的反义寡核苷酸(AODN)抗恶性胶质瘤细胞系的作用.方法 采用脂质体包裹AODN转染恶性胶质瘤细胞系U251-MG细胞.用四唑盐MTT法测定细胞生长曲线,用端粒重复序列扩增法(TRAP)检测细胞的端粒酶活性变化;用流式细胞术检测凋亡细胞.结果 2~8 μmol/L AODN转染恶性胶质瘤细胞系,培养1~4 d,恶性胶质瘤细胞系端粒酶活性下降,且有剂量依赖性和时间依赖性.AODN转染的恶性胶质瘤细胞培养后细胞增生速度减慢,发生了细胞凋亡.而无义寡核苷酸则无此效应.结论 AODN能特异性抑制恶性胶质瘤细胞的端粒酶活性,诱导细胞凋亡.

Abstract: Objective To investigate the effects of telomerase antisense oligodeoxynucleotides(ODNs) on the growth of human malignant glioma cells.Methods Human malignant glioma cells were treated with antisense phosphorothioate oligodeoxynucleotides complementary to the template region of human telomerase RNA(hTR).Cell proliferation was assessed using MTT method.Cell apoptosis was detected by TUNEL.Telomerase activity was detected by telomerase repeat amplification protocol(TRAP) assay.(Results Telomerase) activity and cell growth of glioma cells were suppressed significantly by telomerase antisense oligodeoxynucleotides compared with control group(P<0.05).The proliferation index decreased from 0.46 to 0.25 after 96 h treatment.Telomerase antisense oligodeoxynucleotides also induced cell growth arrest in G0/G1 phase significantly,with reduced cell fractions in S phase and G2/M phase.Results also show that percentage of cells undergoing apoptosis was 10.5 after telomerase antisense oligodeoxynucleotides treatment for 96 h.Conclusion Results suggest that telomerase antisense oligodeoxynucleotides can significantly suppress the telomerase activity,inhibit cell growth and cell proliferation,and induce apoptosis in glioma cells.Telomerase antisense oligodeoxynucleotides are potential therapeutic agents for glioma.
