早幼粒白血病在银屑病病人外周血淋巴细胞中的表达及其与p53和bcl-2家族的相关性

Overexpression of promyelocytic leukaemia gene correlated with p53 and bcl-2 family in psoriatic peripheral lymphocytes

摘要: 目的 研究早幼粒白血病(promyelocytic leukaemia,PML)、p53、B淋巴细胞瘤-2(B-cell lymphoma-2,bcl-2)和bcl-2相关X蛋白(bcl-2 associated X protein,bax)在银屑病外周血淋巴细胞中的表达,并评估上述因子之间的相关性。方法 分离25例银屑病病人和20例健康对照的外周血淋巴细胞,通过qRT-PCR和流式细胞学分别检测PML、p53、bcl-2和bax在外周学淋巴细胞中mRNA和蛋白的表达水平。采用Pearson相关性分析评估PML与上述因子之间的相关性。结果 与健康对照相比,银屑病病人外周血淋巴细胞中PML和p53的mRNA及蛋白表达增加,bcl-2的mRNA及蛋白表达下降,bax的mRNA表达下降但蛋白表达增加。PML蛋白表达与p53、bcl-2以及bax蛋白表达呈相关性。结论 PML可能参与了银屑病发病过程。

Abstract: Objective To investigate the expressions of promyelocytic leukaemia (PML) gene, p53, bcl-2 and bcl-2 associated X protein (bax) in psoriatic peripheral lymphocytes, and evaluate the relationships between PML and those candidate proteins. Methods Twenty-five psoriasis patients and 20 healthy donors were recruited. The peripheral lymphocytes were isolated. The mRNA and protein levels of PML, p53, bcl-2 and bax in peripheral lymphocytes were detected by qRT-PCR and flow cytometry, respectively. Pearson correlation matrix analysis was conducted to evaluate the relationships between PML protein and those candidate proteins.Results The mRNA and protein of PML and p53 were increased, while protein of bcl-2 decreased in peripheral lymphocytes of psoriasis patients as compared to healthy blood donors. The mRNA of bax decreased while protein of bax increased in psoriatic peripheral lymphocytes. The expression of PML protein was correlated with p53, bcl-2 and bax proteins in psoriatic peripheral lymphocytes. Conclusion PML may play a role in the mechanism of psoriasis.
