Abstract: Objective Since its effect of detoxification and laxative, it was found that mast cells and eosinophils have a large concentration on the rat colon after a short-term administration of Rhubarb in our investigation. This phenomenon of allergy caused by Rhubarb has not been reported. The aim of the study is to investigate the function that Rhubarb extract solution should be able to aggrandize mast cells and eosinophils on rat colon. Methods In order to explore the changes of mast cells, eosinophils, interferon-γ (IFN-γ) and interleukin-10 (IL-10) on the effect of different doses of Rhubarb, we gave the rats Rhubarb extract solution by intragastric administration. The samples were measured by means of immunohistochemistry, immunofluorescence and ELISA. Results Rhubarb could make mast cells activated and eosinophils accumulated significantly, causing an increase of IFN-γ and a decrease of IL-10, as while other phenomenon of allergy. Conclusion Administration of Rhubarb for 12 h can cause mast cell and eosinophil enhanced and accumulated in rat colon. It is vague how and why Rhubarb enhances the mast cell and eosinophil, which needs further research.
