人AADC基因的克隆

Gene Cloning of Human Aromatic Amino Acid Decarboxylase

摘要: 为克隆表达人芳香族左旋氨基酸脱羧酶的基因,以了解此基因在多巴胺代谢过程中的作用,从人类胎儿黑质组织中提取总RNA,以RT-PCR方法扩增1442bp的cDNA片段,将此片段克隆于pGEM-T载体中,经过特异性限制性内切酶酶切分析片段正确后,进行全序列分析。结果表明克隆的cDNA与GenBank上的序列相比完全一致,得到了编码正确的人AADC的cDNA全序列。

Abstract: The objective of the study was to obtain the gene of human Aromatic amino acid decarboxylase(AADC), which would be used in gene therapy for Parkinson disease. The fragment of AADC was obtained by RT-PCR from the substantia nigra of humen fetus, and then cloned into the pGEM-T vector and sequenced. Compared with the sequence deposited in GenBank, the sequence of gene cloned in present study is correct.
