摘要:

目的 建立胎儿心脏间充质干细胞(mesenchymal stem cells,MSC)的分离培养方法,并检测其表面标志及多向分化潜能。方法 取胎龄4~5月水囊引产胎儿心脏,用1.073 g/mL的Percoll密度梯度离心分离低密度细胞,MSC培养基纯化贴壁细胞,用流式细胞术检测其表面标志,成脂肪及成骨诱导鉴定多向分化潜能,并与胎儿骨髓MSC对比,研究其增生及生长特征。结果 流式细胞术证明,胎心MSC表达干细胞标志,具有间充质细胞的特征及多向分化能力。原代及传代培养显示,同胎儿骨髓MSC一样,胎儿心脏MSC具有活跃增生的能力。结论 胎儿心脏中可成功地分离MSC,为组织工程的种子细胞和基因工程的载体细胞提供了新的可行的来源。

Abstract:

Objective To isolate and identify the phenotype and biological characteristics of mesenchymal stem cell derived from fetal heart.Methods Mononuclear cells from human fetal heart were collected by Percoll gradient centrifugation.The low density cells were cultivated and expanded in MSC media.Phenotype was analyzed by FACS flow cytometer and characteristics of the multipotent were observed.Results Human fetal heart MSCs were successfully cultivated and differentiated into osteoblast and adipose in vitro as the MSCs derived from fetal bone marrow.Conclusion Mesenchymal stem cells existing in human fetal heart and could be isolated by their adherent ability and should be as a novel source for tissue engineering and gene engineering.
