PABA as Internal Standard in Method of HPLC for Determining 5-FU Concentration in Serum

Abstract

Abstract: To improve the determination of 5-FU by high-performance liquid chromatography, PABA was selected as an internal standard in the analytical method. For determination of 5-FU concentration in serum, a reversed-phase C₁₈ column with a mobile phase consisting methanol-phosphate buffer (pH 6.8) was used. UV detector was set at 270 nm. The limit of determination for 5-FU was 1.2ng. The extraction recovery of 5-FU and PABA were more than 88.1% and 86.55% respectively.
