Detection of Clinical Human Cytomegalovirus Infection from Children by DNA Hybridisation with the Biotinylated Combined Probes

Abstract: A dignostic assay has been developed to detect HCMV DNA in clinical specimens with Biotin-labelled combined probes which were prepared from some cloned fragments of HCMV strain AD169.The labelled probe can detect 25pg of HCMV homologous DNA and fail to hybridize with DNA from other sources.With excellent specificity,the combined DNA probes can detect 100~500 infected cells or 2.57lg TCID50 of HCMV.The assay have high correlation when compared to virus isolation.In the detection of clinical speciments,it correctly identified 3(100%) of 3 urine specimens from congenital deformith positive children.11(61%) of 18 urine specimens from hepatitis positive baby,and 3(75%) of 4 urine specimens from Hormon treatment positive children.There is the evidence that the hybridization assay are specific,sensitive and rapid method for HCMV infection detection.
